PhD position summary/title: PhD Candidate/Student in Design of fusogenic peptides

We’re excited to announce an open PhD student position in Robert Vacha’s Research Group!

Membrane fusion is an essential biological process that plays a crucial role in neurotransmission, intracellular trafficking, and immune responses. Despite its fundamental biological role and potential application in drug delivery, the molecular understanding of membrane fusion and its control remain elusive.

This ERC-funded project aims to the design of novel peptides and peptide aggregates able to induce spontaneous fusion. The peptides have been selected based on their biocompatibility and our exceptional experience with membrane-active peptides, including the design of de novo sequences based on the elucidated mechanism. The first step will be to develop a computational approach to determine the critical peptide properties required to destabilize or stabilize key fusion states: membrane stalk, hemifusion diaphragm, and fusion pore. These findings will then be used to de novo design peptide sequences where the fusogenic role of each amino acid is known, providing the key advantage for customization for vaccination and drug delivery. The computational results will be verified by fluorescence and electron microscopy.

PhD position summary/title: PhD Student in Supramolecular Chemistry

The group of Radek Marek at the Central European Institute of Technology (CEITEC), Masaryk University, is looking for a doctoral student in supramolecular chemistry. If you are passionate about studying the supramolecular properties of anticancer metallodrugs, this is the perfect opportunity for you! In particular, we explore ligand modifications and host-guest complementarity between platinum-based metallodrugs and macrocyclic carriers (cucurbiturils) and their effects on the biological response.

You will have access to a modern chemistry lab and state-of-the-art NMR instruments at our local core facility for regular use. Comprehensive training in supramolecular and NMR techniques will be provided to the successful candidate.

We are seeking a creative, independent, and motivated individual with relevant experience in fields such as structural chemistry, organic chemistry, inorganic chemistry, bioinorganic chemistry. If you have experience in supramolecular chemistry or metallodrugs, we particularly encourage you to apply.

PhD position summary/title: PhD Candidate/Student in effect of lipid asymmetry on membrane protein function

We’re excited to announce an open PhD student position in Robert Vacha’s Research Group!

Eukaryotic and prokaryotic plasma membranes exhibit inherent asymmetry, with distinct lipid compositions between the two leaflets. However, most studies to date have focused on symmetric bilayers. This project addresses this gap by integrating molecular dynamics simulations with experimental techniques such as solid-state NMR spectroscopy and scattering methods.

The primary aim is to elucidate the structural and dynamical properties of lipids in asymmetric membranes, particularly their interactions with integral membrane proteins, with an emphasis on bacterial lipid compositions. Initial step will involve determining lipid chain order parameters, bilayer structure, and lipid dynamics. Subsequently, lipid-protein interactions will be assessed, focusing on lipid-induced modulation of enzymatic activity in two bacterial proteins: OmpLA, a β-barrel protein, and GlpG, an α-helical transmembrane protein. The findings will offer key insights into the role of lipid asymmetry in biological membranes and its influence on membrane protein function, with potential applications in the development of lipid-targeted therapeutics and biosensors for pharmaceutical and biotechnological use.

About Masaryk University, Czech Republic- Official Website

Masaryk University (MU) is the second largest university in the Czech Republic, a member of the Compostela Group and the Utrecht Network. Founded in 1919 in Brno as the second Czech university (after Charles University established in 1348 and Palacký University existent in 1573–1860), it now consists of nine faculties and 35,115 students. It is named after Tomáš Garrigue Masaryk, the first president of an independent Czechoslovakia as well as the leader of the movement for a second Czech university.

In 1960 the university was renamed Jan Evangelista Purkyně University after Jan Evangelista Purkyně, a Czech biologist. In 1990, following the Velvet Revolution it regained its original name. Since 1922, over 171,000 students have graduated from the university.
