PhD position summary/title: PhD Position in Single-Molecule Biophysics using Nanopore Trapping
Our group has pioneered the development of the Nanopore Electro-Osmotic Trap (NEOtrap), a groundbreaking technique that enables label-free trapping and sensing of single proteins in solution for up to hours. Now, we leverage the unique abilities of nanopore trapping to detect proteins and their conformations, dynamics, interactions, catalytic mechanisms etc. in real-time. Building on our published and unpublished work, the successful candidate will advance nanopore trapping in new directions using solid-state and biological nanopores to identify previously inaccessible protein properties with single-molecule resolution, for future applications in biotechnology and biomedical diagnostics.

(02) PhD Degree – Fully Funded
PhD position summary/title: PhD Position in Single-Molecule Biophysics using Fluorescence & DyeCycling/FRET
Biomolecular dynamics, such as conformational changes, are the understudied link between biomolecular structure and function. Single-molecule FRET is an established technique, unique in its capacity to resolve nanometer small conformational changes within single biomolecules (proteins, DNA, RNA), without suffering from ensemble averaging. Its Achilles heel is, however, photo-bleaching of the fluorescent dyes involved, which ends the experiment prematurely, rendering many biological questions inaccessible. To bypass this limitation, our group has developed DyeCycling/FRET, where the dyes are continuously replaced. Building on our published and unpublished work, the successful candidate will advance nanophotonic and fluorogenic DyeCycling to map protein energy landscapes in unprecedented detail, thereby gaining access to previously overlooked effects, like dynamic disorder, with far-ranging consequences for the design and functional understanding of protein systems.

The successful PhD applicant will develop a PhD research program in the research fields listed above, or a closely related field, in close collaboration with the daily supervisor Prof. Dr. Oliver Heiri and the Geoecology group. The research should contribute to methodological and interpretational advancements in the field of chironomid palaeoecology by applying new methods to fossil chironomid research, applying new approaches for interpreting fossil chironomid records or developing new types of calibration data for further expanding the use of aquatic invertebrate and particularly chironomid remains in lake sediment studies. The successful candidate will also contribute to other research and teaching activities of the Geoecology group and is expected to collaborate closely with other research group members and researchers from other research institutions.

PhD position summary/title: PhD student in Molecular Toxicology
The project is funded by the Swiss Centre for Applied Human Toxicology (SCAHT). This interdisciplinary research project is a collaboration with four partner institutions and focuses on investigating disruptions of steroidogenesis induced by endocrine-disrupting chemicals (EDCs). EDCs are exogenous substances or mixtures interfering with hormone receptors and signalling pathways or altering hormone synthesis, transport, and metabolism, ultimately leading to adverse health effects. Steroid hormone producing glands such as the adrenals, ovaries and testes are particularly susceptible to EDCs due to their highly vascularized, lipid-rich environment necessary for steroid biosynthesis, and the presence of cytochrome P450 enzymes that produce free radicals and reactive intermediates. These properties make steroidogenesis a critical target for chemical-induced endocrine disruption. By disturbing steroid biosynthesis EDCs are posing risks for metabolic, immune, neuronal and long-term reproductive diseases in both males and females. While compounds such as phthalates and bisphenol A (BPA) have been well studied, there is limited knowledge about the endocrine-disrupting potential of their newer derivatives and replacement chemicals. Addressing this knowledge gap is essential for identifying potential hazards posed by such emerging substances. The goal of this project is to assess the impact of phthalates, bisphenols, and more importantly their analogues on steroid hormone biosynthesis in adrenal, testicular, and ovarian systems. The study will primarily employ in vitro cell-based assays to evaluate these effects. Depending on activity and relevance from the literature, the most promising active compound(s) may be further investigated in animals.

PhD position summary/title: PhD position in alpine and arctic plant ecology
We are looking for a highly motivated candidate with a self-organized and solution-oriented work attitude. Strong communication and organizational skills, the ability to work in teams and a commitment to a collegial and inclusive workplace are essential. Applicants should hold a MSc degree in ecology, botany, biogeography, environmental science, computational biology or related fields. Very good written and spoken English is a must.
