PhD position summary/title: Phd Position in computational biology (Zampieri Lab)
The mission of the group is to understand fundamental mechanisms regulating short- and long-term metabolic adaptation to genetic and chemical perturbations to find new and unconventional therapeutic strategies, ranging from antibacterial to anticancer drugs. To this end, we develop new ways to combine state-of-the-art technologies in metabolomics with mathematical modeling.
The candidate will have the opportunity to exploit some of the cutting-edge experimental and computational methods, comprising constraint-based and kinetic modeling, statistical analysis of large datasets, high-throughput metabolomics, time-lapse microscopy, to investigate how to pharmacologically interfere with fundamental mechanisms in the regulation of bacterial metabolism. Available resources at the department include a mouse facility, high-end FACS, IT and microscopy facilities, the Life Sciences Training Facility (for gene expression and proteome profiling) and much more.
The candidate is expected to have a strong background and interest in quantitative disciplines, excellent teamwork and communication skills in English. The candidate will have the opportunity to develop the following project with a lot of academic freedom and strong support from senior members in the lab, and at the same time to play an active role in shaping and in creating inspiring research and working environment.

PhD position summary/title: PhD student position: Biochemistry/Molecular Toxicology
Corticosteroids regulate essential physiological processes. Substances altering their production, cell-specific metabolism or receptor-mediated signalling can aggravate various diseases including inflammatory, cardio-metabolic as well as developmental and reproductive disorders. Despite the high interest in identifying endocrine disrupting chemicals (EDCs), the identification and characterization of substances disturbing corticosteroid action and elucidation of their mechanism-of-action is a largely neglected topic.
The present project is a collaborative effort with four other partners to study chemical-induced steroid perturbations. The project is financed by the Swiss Centre for Applied Human Toxicology. The aim of the project is to establish and apply methods and cell-based assays to assess chemical-induced alterations in glucocorticoid pre-receptor enzyme activity and regulation. Promoter-reporter cell lines will be generated for HSD11B1, converting inactive cortisone to the potent glucocorticoid cortisol, and HSD11B2, catalysing the reverse reaction. Enzyme activity assays that are already established will be applied to test for direct inhibitory activity of compounds. Cell lines stably expressing either these promoter-reporter constructs or glucocorticoid receptor (GR) reporter plasmids will be constructed. Cell lines of interest include placental JEG3, colon SW620, and renal RCCD2 for HSD11B2, and hepatic and macrophage cell lines for HSD11B1. Compounds that will be included in the analyses comprise PFAS, phthalates, bisphenols, UV-filters and parabens. Depending on activity and relevance from literature, the most promising active compound(s) can be further investigated in additional cell models or in animals. Methods applied include cell culture, transfection and gene expression analyses, molecular cloning, receptor function analyses, and enzyme activity measurements.

- Study the behavior of cells seeded on novel materials focusing on biocompatibility, tissue phenotype, and matrix deposition.
- Development of an osteoarthritic model based on a paper-microfluidic device
- Study of drug diffusion and cellular responses in real-time using electrochemical sensors

The Faculty of Psychology is one of seven faculties of the oldest university in Switzerland. With our core focus on Society & Choice and Health & Interventions, we are successful both nationally and internationally in our research and study programs.
In the newly established research group “Statistics & Data Science”, we develop and evaluate statistical models and analysis methods in psychology. Our research focuses on psychometric models (e.g., Item Response Theory models), machine learning methods (e.g., Random Forests, Neural Networks, and interpretation techniques), and multilevel models (e.g., analysis methods for hierarchical and longitudinal data).
