Ord är centrala för mänsklig kommunikation och identitet. För personer med afasi eller demens kan svårigheter att hitta rätt ord påverka både självständighet och livskvalitet. Detta doktorandprojekt syftar till att fördjupa förståelsen för hur ordsökningsproblem tar sig uttryck i olika sammanhang och hur dessa påverkar interaktionen.

Projektet omfattar fyra delstudier och bygger på testdata samt videoinspelade samtal och språktester med totalt 40 deltagare (20 med afasi och 20 med demens), samt deras närstående. Genom multimodal interaktionsanalys undersöks hur kroppsspråk, gester och andra icke-verbala resurser samspelar med talet i kommunikationen. Dessa resultat jämförs med kvalitativa och kvantitativa analyser av testresultat.

PhD position summary/title: Doctoral position in health data science

Hydroclimate extremes—such as floods, droughts, and heatwaves—have severe consequences for both human and natural systems, contributing to over 2 million deaths globally in the past 50 years. Climate change is increasing the frequency and intensity of these events. Infants are among the most exposed and vulnerable groups, highlighting the urgent need to reduce their risks through improved recommendations and long-term mitigation strategies.

This PhD project aims to develop an innovative modelling framework to quantify the societal and health impacts of hydroclimate extremes on early life, while also considering adaptation measures. The specific objectives are to:

  • Quantify the societal and health impacts of hydroclimate extremes and climatic factors during early life;
  • Apply Distributed Lag Non-Linear Models (DLNMs) to assess the delayed and non-linear effects of hydroclimate extremes and climatic factors on early-life health and social outcomes.

The PhD student will contribute to study design and data analyses using a data science approach, and the interpretation and presentation of results through scientific publications and conference presentations.

(03) PhD Degree – Fully Funded

PhD position summary/title: Doctoral (PhD) student position in immunology and adoptive cell therapy

Adoptive Cell Therapy (ACT) with tumor infiltrating lymphocytes (TIL) has successfully ben used as therapy for patients with malignant melanoma and is now practiced at a number of clinical centers worldwide, including at Cancer Center Karolinska (CCK). At CCK we have the only GMP unit in Sweden with expertise and the authorization to use TIL therapy to treat patients with metastatic melanoma.

Our ACT clinical trial is unique as it is combined with a Dendritic cell (DC) based tumor vaccine. The results are encouraging and shows that the combination of TIL cells and a DC vaccine induce improved clinical responses. The overall aim is to build on these clinical results and improve ACT for patients with metastatic melanoma and furthermore extend these findings to other solid cancers focusing on gynecological cancers.

The candidate will have several projects aiming to improve and refine the cell product used to treat patients with a metastatic disease to achieve a better anti-tumor response and a better clinical outcome. This will include projects involving projects investigating the efficacy of combining TIL and an oncolytic virus to enhance tumor elimination of ovarian cancer.

PhD position summary/title: Doctoral (PhD) student position in probabilistic models of proteins

This Data Driven Life Sciences (DDLS) PhD project focuses on probabilistic models of protein structure, which can be used primarily for computational protein design, but also for an array of other applications related to modeling and understanding proteins and their behavior. The heart of the project will be the development and refinement of novel computational methods and models, including extending methods already under development in the lab, with a particular focus on ways of exerting more precise control for protein design.

In collaboration with the co-supervisors and their labs, the project will also involve the use of these methods (and methods developed by others) for applications in infection biology, chiefly for designing immunogens for new vaccines, but also exploring other applications. Another aim is to use the properties specific to some of the methods we are developing to try and improve secondary model use cases, such as evaluating designed backbones and predicting the functional effects of protein variants.

I forskningsprojektet ”Säkerhet och effektivitet av behandlingar för immunmedierade inflammatoriska sjukdomar: är datadrivna modeller för kausal inferens i praktiken bättre än traditionella metoder?” vill vi utforska hur olika nya förslag på statiska metoder som kombinerar nya möjligheter för datadriven forskning genom maskininlärningsalgoritmer med teoretiskt utvecklade modeller för kausal inferens påverkar resultaten i en rad av kliniskt relevanta situationer, jämfört med olika mer etablerade modelleringsalternativ. Genom att blanda studier i äkta data för en rad sjukdomar (främst multipel skleros och reumatoid artrit) och utfall med simuleringsstudier belyser vi dels hur metoderna fungerar och kan användas i praktiken, och utforskar när de teoretiska fördelarna i praktiken påverkar slutsaterna från studien, och därmed rekommendationerna för klinisk praxis. Doktoranden förväntas vara drivande i samtliga studier, stå för all programmering, stödja insamlandet av klinisk data, samt presentera sin forskning i vetenskapliga publikationer och vid konferenser.

PhD position summary/title: Doctoral (PhD) student position in skin stem cell biology

The PhD project focuses on understanding the cellular origins of skin. How, when, and why do cells commit during development to form the vast repertoire of specialized skin cells? The aim of the PhD program is to determine the developmental connectivity of skin cells, such as keratinocyte and fibroblast subtypes, from early development through morphogenesis to adult skin. A combination of experimental and computational state of the art techniques will be used, such as in vivo clonal barcode tracing, single-cell RNA/ATAC -seq and spatial transcriptomics, and computational biology.

The PhD program at the Karolinska Institutet corresponds to 4 years full time studies. The project is a combination of practical laboratory work (wet-lab work, imaging, mouse work) and computational multi-omics single-cell data analysis. The studies also include attending seminars, courses, and conferences. The PhD student will under supervision, and with time more independently, plan and execute experiments, analyze data, interpret, and present the results as well as write scientific articles.

Projektets övergripande mål är att förstå hur förändringar i lysosomnivåer påverkar cancercellers metabolism, tumöraggressivitet och behandlingsresistens. Forskningen fokuserar särskilt på medulloblastom, en aggressiv barnhjärntumör där återfall ofta innebär en mycket dålig prognos. I projektet studeras hur förhöjda lysosomnivåer kan driva tumörprogression och hur detta påverkar centrala tillväxtsignaler i cellen, såsom mTORC1-signalering. Dessutom undersöks de molekylära mekanismer som reglerar organisationen av det lysosomala nätverket i celler. Projektet kombinerar avancerade metoder inom cell- och molekylärbiologi, inklusive qPCR, cellodling, konfokalmikroskopi och biokemiska analyser. Som doktorand i detta projekt kommer du att vara aktivt involverad i alla delar av forskningsprocessen – från hypotesgenerering och experimentell design till praktiskt laboratoriearbete, dataanalys, manusförfattande och presentation av forskningsresultat vid nationella och internationella konferenser.

Som doktorand kommer du att undersöka biomarkörer och patientrapporterade utfallsmått för klinisk förbättring och behandlingssvar hos patienter med den reumatiska systemsjukdomen myosit. Studierna kommer främst att vara epidemiologiska och baseras på data från två av världens största myositregister. PhD position summary/title: Doctoral (PhD) student position in Systems immunology

This project is about studying and modulating the function and phenotype of immune cells using microbial contacts and stimulation with ligands. We want to analyze how the function of white blood cells is regulated using protein distribution on the cell surface. We believe that such regulation affects the function of immune cells dynamically and want to study this and try to influence protein localization in immune cells using drugs such as bispecific antibodies etc. Our hypothesis for the project is that protein distribution, which can be read out with Pixelgen’s new methods (MPX), can tell us something about the regulation of immune cells in disease such as inflammatory diseases and in health during different phases of life.

PhD position summary/title: Doctoral (PhD) student position in molecular chaperone-amyloid interaction for biomimetic treatment of neurodegenerative diseases

This PhD project concerns the study of amyloid formation in neurodegenerative disorders, such as Alzheimer’s disease and Parkinson’s disease, and the implication of molecular chaperones, in particular the BRICHOS domain. BRICHOS has excellent properties as an anti-amyloid molecular chaperone, inhibiting amyloid formation and associated toxicity and is hence a promising candidate for novel treatment approaches.

The aims of the project are (i) to better understand the molecular mechanisms of Tau protein aggregation associated to Alzheimer’s disease and (ii) to identify the mechanism-of-action of chaperone-amyloid recognition to establish a basis for development of future therapeutic strategies against neurodegenerative disorders.

This PhD project is multidisciplinary and combines protein biochemistry and structural biology in the context of neurodegenerative diseases. For the PhD student this project offers excellent opportunities to perform cutting-edge research in a friendly and inspiring work environment, learn state-of-the-art biochemical and biophysical techniques with the possibility to contribute to the search of new protein-based therapeutics. In particular, cryo-electron microscopy in combination with solid-state and solution nuclear magnetic resonance will be applied in the project.

The PhD student will perform major parts of the experimental work of this project and will plan the experiments, analyze the data and interpret and summarize the results. The work will be conducted both independently and together with other team members, and the PhD student is encouraged to develop own ideas. Courses of 60 credits related to the research topic are also included in the doctoral project, as well as participating in supervision of undergraduate students and attending international scientific conferences.

PhD position summary/title: Doctoral (PhD) student position in CRISPR functional genomics of cancer drug resistance

Drug resistance is a major obstacle to curative cancer therapy. Building on our recent discovery of metabolic stressors that suppress tumor growth and expose novel vulnerabilities, the PhD project will:

  • Dissect how metabolic stress reshapes drug responses and resistance evolution in e.g., BRAF‑mutant cancer cells treated with precision drugs.
  • Run genome‑wide CRISPR screens to pinpoint genes and pathways that modulate resistance, followed by validation in patient‑derived samples and mouse models.
  • Assess the impact of metabolic stress on the immune system in general and on anti‑tumor immunity more specifically.

The student will master cell culture, molecular biology, flow cytometry, next‑generation sequencing, in vivo tumor models, and computational analysis, and will present results in international conferences and first‑author publications.

PhD position summary/title: PhD student position in Molecular mechanisms of drug action

There is currently a large knowledge gap as to why males and females respond differently to treatments for metabolic disorders. This is due, in large part, to the lack of sex-balanced studies in metabolic research. Most studies leave out females due to hormonal variability and differences in fat storage and energy metabolism. Several metabolism-related disorders, including type 2 diabetes and metabolism-associated steatotic liver disease (MASLD), exhibit notable differences in disease prevalence and progression between men and women. Focusing on important targets for drug development, this PhD project aims to uncover the sexual differences in G protein-coupled receptor (GPCR) signalling using patient-derived organotypic models.  

To achieve this central aim, the PhD candidate will employ a battery of assays with a focus on cell and molecular biology (2D and 3D tissue models, phenotypic assays, signal transduction assays, omics-data generation), bioengineering (organ-on-a-chip system) and bioinformatics (omics-data analysis). By the end of this project, lead molecules with novel signalling properties will be developed to address existing hurdles in sex dimorphism in diseases like MASLD.

The student is expected to play a strong role in hypothesis generation, experimental design, experimentation, data analysis, presentation of results at meetings and conferences, and manuscript preparation. The student will be responsible to follow the general rules and regulations and time plan of the doctoral program.
