Within your postdoctoral studies you will study a range of novel ASO-conjugates for their ability to silence genes in cell culture and in mice models, with focus on heart delivery. Firstly, fluorescent ASOs conjugated to a range of ligands will be studied for cellular uptake in different cell lines, including cardiomyocytes, using flow cytometry and confocal microscopy. Subsequently, dose-and time response studies will be performed using qPCR to determine the magnitude of gene silencing with the different conjugates across different cell types. Next, in vivo biodistribution studies will be performed using IVIS imaging, following intravenous or subcutaneous administration. Plasma will additionally be collected for PK analysis and to study protein adsorption. Lead conjugates will then be tested for functional silencing of target genes in different organs in mice following systemic administration of ASOs. To determine the magnitude of silencing in individual cells, different methods will be employed. In screening experiments, the non-coding RNA Malat-1 and the membrane protein CD47 will be used as targets. These are chosen as they are both ubiquitously expressed body-wide but reside in different cellular compartments. CD47 silencing will be assessed by flow cytometry and immunohistochemistry analysis together with antibodies for staining of specific cell types. Malat-1 silencing will be analyzed using single cell RNA sequencing of cells that have taken up Cy3-labelled ASO conjugates in vivo. Using this methodology, it is possible not only to determine the knockdown levels but also call the identity of the cell types within an organ that have internalized the conjugates. In this way, it is possible to distinguish productive uptake from mere cellular internalization into individual cells in vivo.

In cancer, targeting the immune system is a promising therapeutic approach. But immunotherapy only benefits a fraction of cancer patients because tumor immunology is largely patient-specific. Designing immunotherapy effective in a given patient is challenging because anti-cancer immunity is determined by the interaction of many molecules and cells according to laws that are only partially and imperfectly understood. To reveal these laws, our lab takes advantage of technological advances that quantify thousands of molecules in thousands of tumor cells through cancer progression and therapy. Using this data, we develop physics-inspired AI approaches to model how molecules and cells determine the success or failure of anti-cancer immune response. The vision is for these models to guide immunotherapeutic intervention in the clinic.

The successful candidate has a background in, and previous experience with, computationally intense methods/ML/prediction modeling, through applications during a PhD or MSc. Previous work experience with medical or biological data is a requirement, so is experience of real-world data, data management, and epidemiological studies. You have formal training in, and experience of, statistical analyses. The candidate is proficient in programming languages such as R, SAS and Python, and has experience of parallelization of code. The successful candidate enjoys working in an interdisciplinary team, is thorough and organized, and structures and documents well. Within the department and KI, you will be interacting also with clinical and translational RA researchers. As a Postdoctoral Research fellow, you will be included in the work on the design and synthesis of synthetic ligands and improved strategies for bioconjugation of ligands to ASOs and siRNAs. Most of the research will exploit methods used in general organic chemistry laboratories including: synthesis, analysis and purification strategies (Flash column chromatography, MS/NMR spectroscopies, HPLC purifications). The duties will additionally involve synthesis of peptides and ASOs using solid phase approaches, their modifications and bioconjugations to various entities. The methods used for bioconjugation include amide bond formation, a number of click chemistry approaches, solid support functionalization and more.

We are seeking a highly motivated postdoctoral researcher with a background in neuroscience to join our team. The primary objective of our research is to characterize circadian processes involved in both metabolic and mental disorders. As a successful candidate, you will lead a neuroscientific branch in our lab and collaborate closely with researchers specializing in metabolism.

Both candidates must hold a PhD in immunology, rheumatology or equivalent. Moreover, fluency in english is a requirement. The first project will require expertise in cell culture of primary cells and multiparameter flow cytometry. We have a long-standing interest in basic and clinically relevant aspects of unconventional T cells (UTC), such as mucosal associated invariant T (MAIT) cells, invariant natural killer T (iNKT) cells or ɣẟ T cells. The main task of the postdoctoral fellow will be expand the current knowledge on a novel subset of UTC known as CD4- CD8- ⍺β T (DNT⍺β) cells in humans, in the context of homeostasis and during inflammatory states. The successful applicant shall be driven by a strong ambition to independently develop and lead the project within the defined scientific area. The project success shall rely on solid theoretical bases on UTC immunology and on existing skills with several technological platforms, including high-dimensional flow cytometry, cell sorting and single cell transcriptomics.

We search for a post-doctoral fellow who can work together with Professor Annelie Brauner and her team within a project of innate immunity with focus on the host-microbe interaction with special focus on E. coli infections in the urinary tract.
 Infection is a vast individual and socio-economic problem especially because of the increasing antibiotic resistance, which makes antibiotic treatment increasingly difficult. Among infections, urinary tract infections, often caused by E. coli belong to the top 10 infections, mainly in women and children. Interestingly, persons with underlying diseases such as diabetes mellitus have increased risk of infections and frequently have recurrent urinary tract infections and also severe infection complications. Such complications are sometimes life threatening. From a global perspective, urinary tract infections are still a main cause of renal insufficiency. Based on the worldwide increasing antibiotic resistance new treatment strategies are urgently called upon.

The postdoc position focuses on inflammatory and carcinogenic effects induced by silica and related chemicals. We work with cell models and study cell signaling at a molecular level. The aim is to understand aspects of silica-induced DNA damage and its possible influence on carcinogenesis. Included are studies on DNA damage signaling and other events of possible importance for carcinogenesis, including NALP3 activation. The applicant should have a PhD degree from an appropriate discipline, such as cell- or molecular biology or toxicology. Experience in cell- and molecular biology techniques is needed. Experience in such techniques of relevance for carcinogenesis and inflammation is a merit. The position is temporary for 2 years, with possibility of extension.
