At Scripps Research, the Lotz Laboratory is studying mechanisms and treatments of intervertebral disc degeneration (IDD) which affects a large population of older individuals and represents the major cause of back pain.  The research on disease mechanisms uses human spines and genetic mouse models to investigate changes in cells, genes and proteins that develop during the aging process and the development of IDD. These studies examine cell function, RNA expression patterns using RNA sequencing and single cell RNA sequencing.

Postdoc summary/title: POSTDOCTORAL ASSOCIATE – Molecular Medicine (RE-JOIN) – Lotz Lab

The Dr. Lotz lab at Scripps Research is looking for a highly motivated individual with strong background and technical expertise in cell and molecular biology and bioinformatics for our program on transcription factors in joint biology and arthritis pathogenesis. In a newly funded program about mechanisms of joint tissue destruction and joint pain, we are using Omics technologies (RNA-seq, single cell-RNA-seq, ChIP-seq, epigenetic analyses, spatial transcriptomics) to identify dysregulated transcription factors and test candidate genes in vitro and using mutant mouse strains. Genes that are identified to be essential regulators of homeostatic processes or to promote arthritis pathogenesis are tested in viral gene delivery approaches and used in drug screening assays.

(03) Postdoctoral Fellowship Position

Postdoc Fellowship Position summary/title: POSTDOCTORAL ASSOCIATE – Teijaro Lab

Postdoctoral Associate position is available immediately at The Scripps Research Institute in the laboratory of Dr. John Teijaro, whose lab is interested in studying the innate and adaptive immune response to acute and persistent viral infections. Immune suppressive environments during persistent/chronic viral infection prevent excessive immune pathology but also impede the ability of the immune system to eradicate infectious virus from the host. Conversely, excessive inflammation during acute viral infections can reduce viral loads but also lead to severe immune pathology and even death. The focus of the Teijaro Lab is understanding how immune suppressive and pathological environments are generated, maintained and reversed, with a specific focusing on how type 1 interferon (IFN-I) production and signaling regulates these two states. The Teijaro lab is looking at various viral infection models with respect to the above including: LCMV, Influenza, SARS-CoV2 and Measles virus.

(09) Postdoctoral Fellowship Position

Postdoc Fellowship Position summary/title: POSTDOCTORAL ASSOCIATE – Paust Lab

The research projects will focus on NK cell responses to infection and malignancy, using mice, humanized mice, and human specimens.

Project 1. To develop memory NK cell-based immunotherapies to prevent or cure HIV infection.  This work will be performed using in-house generated humanized mice with a competent immune system and human specimens.

Project 2. To develop NK cell-based immunotherapies to prevent or cure solid tumors. This work will be performed using genetically engineered lung cancer mouse models and in-house generated patient-derived xenograft models (lung, and pancreatic cancer).

Project 3. To understand NK cell responses to respiratory virus infection (mainly Influenza virus) in the context of innate and adaptive immune crosstalk and immune-therapeutics development. The work will be performed using mouse models of influenza virus infection and in vitro assays.

The Ghadiri Laboratory at the Scripps Research, Department of Chemistry, and the Skaggs Institute for Chemical Biology is currently seeking talented Postdoctoral Associate candidates with a strong background in biology, molecular biology, chemical biology, and/or biochemistry. The lab is developing new, general approaches to control the site and timing of enzyme activity. Enzymes have great potential as therapeutic and diagnostic agents due to their high specificity and catalytic activity, but the ability to better control the timing and location of enzyme function would enable the use of a wider variety of enzymes for therapeutic purposes. Our approach for enzyme programming relies on attaching an enzyme to its small-molecule inhibitor through an allosteric DNA tether. Upon interaction of the DNA tether with its complement strand, the ensuing conformational change activates the enzyme by dislodging the inhibitor from its active site. We have established the utility of DNA-programmed intrasterically-regulated enzymes in developing constructs for fast, sensitive, and PCR-independent isothermal detection of label-free nucleic acid sequences; carrying out complex logic operations; programming the activity of streptokinase, a clinically-used fibrinogen activator; and specifically killing targeted cancer cells.
