(18) Postdoctoral Fellowship Position

Postdoc Fellowship Position summary/title: Postdoctoral Researcher/The Strategic Research Programme for Diabetes (5)

We recently demonstrated that glutamine levels are lower in white adipocytes from obese compared to lean subjects and that this leads to white adipose tissue (WAT) inflammation (Petrus, Lecoutre et al, Cell Metabolism, 2020). Our new preliminary data in human and murine WAT, reveal that these changes are associated with decreased expression of Glutamine Synthase (GS, encoded by GLUL) and increased expression of glutaminase (GLS, encoded by GLS), the two enzymes governing glutamine metabolism. This suggests that glutamine turnover is a major determinant of adipocyte function. To further dissect the pathophysiological role of our observations in vivo, we have generated adipocyte-specific Gls and Glul knockout mice where we will study the effects of different perturbations on WAT and whole body metabolism. These analyses will be complemented by state-of-the-art mechanistic studies in human adipocytes in which we for instance will perform metabolic flux analyses following CRISPR/Cas9-mediated engineering of the glutamine pathway. Our project will allow the applicant to work with an array of methods in clinical translational research and provide unprecedented insights into the immunometabolic role of adipocyte glutamine turnover.

Postdoc Fellowship Position summary/title: Postdoctoral Researcher in NK cell biology

The Malmberg Lab is located in Stockholm/Sweden and Oslo/Norway and currently consists of 24 people. We are now looking for a highly motivated post-doc with an interest in molecular immunology and experimental immunotherapy. The project concerns the spatiotemporal regulation of NK cell education with a particular focus on the cellular and molecular mechanisms that allow NK cells to translate receptor input into a “molecular memory” and how this is affected in the tumor microenvironment.  The studies involve basic molecular immunology with a strong translational component and is carried out in close collaboration with our industrial partner Merck and the Malmberg Lab at Oslo University Hospital. One goal is to develop in vitro assays to study NK cell tuning and use a variety of single-cell assays, including high-dimensional immune profiling by flow cytometry, single-cell indexed FACS-sorting and RNA Seq. The project also involves molecular biology/cloning, transient transfection / overexpression, retroviral and lentiviral transduction in mammalian cells. Additional tasks include gene-editing of primary NK cells / NK cell lines by CRISPR/Cas9, cytotoxicity assays, and qPCR. The postdoctoral fellow is expected to plan, execute, and analyze experiments in an independent manner and contribute scientifically to the group.

Postdoc Fellowship Position summary/title: Postdoctoral studies in Central Nervous System Regeneration after Spinal Cord Injury (scholarship)

Our lab investigates stromal scar formation and its impact on axonal regeneration and functional recovery following spinal cord injury. We discovered a new population of perivascular cells, called type A pericytes, as the source of fibrotic scar tissue (Göritz et al., Science, 2011) and identified pericyte-derived scar formation as a therapeutic target to achieve axonal regeneration and functional recovery following spinal cord injury (Dias et al., Cell, 2018). Recently, we showed that fibrotic scarring by perivascular cells is a conserved mechanism in the adult mammalian CNS (Dias et al., Nature Com., 2021). The intended project will test different strategies to interfere with fibrotic scarring to enhance axonal regeneration and functional recovery following spinal cord injury.

(25) Postdoctoral Fellowship Position

Postdoc Fellowship Position summary/title: Postdoctoral Researcher/The Strategic Research Programme for Diabetes (3)

Comorbidities of type II diabetes (T2D) are major health concerns worldwide, including peripheral artery disease (PAD). PAD lowers blood flow to the lower limb, causing muscle ischemia and dysfunction and reduced exercise tolerance. Functionality of limb skeletal muscle is a key component for determining morbidity and mortality risk in patients with PAD. Interventions that improve distal arterial pressures (ie. bypass surgery) generally fail to normalize the structural and functional abnormalities of muscles, which point towards pathophysiological mechanisms inside the skeletal myofibers that reduce overall muscle function. Despite serious muscle abnormalities, the field is lacking molecular understanding of PAD-induced muscle dysfunction. Here we aim to use a translational approach involving PAD patients and mouse models and state-of-the-art methods going from functional analyses to single cell profiling to illuminate molecular mechanisms underlying PAD-induced muscle pathophysiology. In the project, human skeletal muscle biopsies, as well as muscle from preclinical model, will undergo multiple assessments e.g., global transcriptomic analyses and single-cell profiling, histopathological analysis (electron microscopy, immunofluorescence), mitochondrial respiration, and contractility (force) measurements. Muscle cultures, isolated muscle fibers and in vivo models will be used for gain- or loss-of-function experiments to assess selected molecular candidates from the transcriptional analyses.  The expected outcome of this curiosity-driven project is detailed molecular understanding of intramuscular signaling and contractile function, which will yield substantial novel insights into PAD muscle pathology and identify targets for future therapeutic development.

About The Karolinska Institute, Sweden – Official Website

The Karolinska Institute sometimes known as the (Royal) Caroline Institute in English is a research-led medical university in Solna within the Stockholm urban area of Sweden. The Nobel Assembly at the Karolinska Institute awards the Nobel Prize in Physiology or Medicine. The assembly consists of fifty professors from various medical disciplines at the university. The current rector of Karolinska Institute is Ole Petter Ottersen, who took office in August 2017.

The Karolinska Institute was founded in 1810 on the island of Kungsholmen on the west side of Stockholm; the main campus was relocated decades later to Solna, just outside Stockholm. A second campus was established more recently in Flemingsberg, Huddinge, south of Stockholm.

The Karolinska Institute is Sweden’s third oldest medical school, after Uppsala University (founded in 1477) and Lund University (founded in 1666). It is one of Sweden’s largest centres for training and research, accounting for 30% of the medical training and more than 40% of all academic medical and life science research conducted in Sweden.

The Karolinska University Hospital, located in Solna and Huddinge, is associated with the university as a research and teaching hospital. Together they form an academic health science centre. While most of the medical programs are taught in Swedish, the bulk of the PhD projects are conducted in English. The institute’s name is a reference to the Caroleans.
