Postdoc Fellowship Position summary/title: Postdoc position in Primary cilia and pH-regulatory ion transport proteins
Most cells in our body contain antenna-like structures on their surface called primary cilia, which are important for regulating cell-cell communication and thereby the development and function of our organs. Defects in cilia can lead to severe human diseases called ciliopathies, which often affect the kidneys. The aim of this project is to investigate how two pH-regulatory ion transporters, NBCn1 and NHE1, are targeted to the primary cilium of mammalian kidney cells, and how they regulate pH within the cilium. NBCn1 and NHE1 regulate essential processes in our cells and organs and their dysfunction is linked to diseases like cancer and hypertension, but it is not clear how they get localized to and function at cilia. Likewise, intraciliary pH and its possible effect on ciliary function are unknown. The results will provide new insight into fundamental cellular trafficking mechanisms, the regulation and subcellular functions of NBCn1 and NHE1, and into how pH may affect cilia-mediated signaling.

The project will address transmission of chromatin organization trough DNA replication and how this is linked to genome/epigenome stability, including the interplay with replication timing. The interplay of histone dynamics and chromatin replication with epigenome and genome stability remain poorly understood. Our recent discovery that new histones control repair pathway choice and the identification histone recycling mechanisms are new entry points to these questions (Becker et al., 2021 Nature; Nakamura et al., 2019 Nat Cell Biol; Petryk et al., 2018 Science, Saredi et al., 2016 Nature). You will join a multi-disciplinary research team working on how epigenetic information is transmitted to daughter cells and guide cell fate choices. The project aims at developing imaging method to achieve multichannel super-resolution imaging with conventional fluorescence microscopy. Recently, we have achieved a breakthrough by developing the brightest full-organic fluorescent nanoparticles based on a molecular self-assembly approach, called: Small‐Molecule Ionic Isolation Lattices (SMILES)*. Besides their unsurpassed brightness, we also have the full control of the fluorescence emission color (from blue to near infrared) and fluorescence lifetime (from nanosecond to microsecond). In this project, we will extend the number of simultaneous detection channels and develop multiplex microscopy by combining the “normal” color channels with fluorescence lifetime channels imaging. We will use the developed multiplexed imaging method to tackle the key question about protein translation at the subcellular level.

The postdoc position covers structural cell biology and will be based in the Plant Cellulose Synthesis research group in a highly cross-disciplinary team that includes the full range of computational structural biology, microscopy, molecular biology and biochemistry (> 20 people). This postdoc will prepare single cell cultures (protoplasts) derived from plant tissue of transgenic marker lines (A. thaliana and/or Nicotiana tabacum BY-2) and localize the CSC complex in vivo using correlative light and electron microscopy (CLEM) through the group’s access to microscope facilities e.g. Center for Advanced Bioimaging (CAB) and Core Facility for Integrated Microscopy (CFIM) both located at the University of Copenhagen. You will take part in the design and execution of experiments and measurements in laboratories and in the field. GPCRs are the targets of around 30% of approved drugs and development of new and improved therapies is a major focus of academic groups and companies worldwide.Chemokines drive cell migration through interactions with chemokine receptors, thereby controlling the immune system, development and other functions. Chemokine receptors and other GPCRs are dynamic proteins and regulated by a number of cellular factors including membrane lipids. We have identified specific lipids with different functional effects on chemokine receptors. The project aims to characterize the molecular mechanisms of lipid regulation using a combination of structural and functional experiments in cells and reconstituted systems. Your tasks will include expression, purification and reconstitution of receptors and characterization of receptor structure, dynamics and function.
