Postdoctoral position summary/title: Postdoctoral research fellow – Translational studies on the sarcoma tumor microenvironment

The successful candidate will perform explorative studies to map molecular and cellular communication between different cell types in sarcoma. Tumor-suppressive versus tumor-promoting functions of immune cells, mesenchymal stromal cells and blood vessels will be explored at the molecular, cellular and tissue level in vitro and in vivo. (25) Postdoctoral Position

Postdoctoral position summary/title: Postdoctoral studies – macrophages in cardiometabolic disease (scholarship)

Cardiometabolic disease is to a large part underlying the vast increase in non-communicable diseases observed in recent years. Particularly relevant for the current project are the components non-alcoholic fatty liver disease (NAFLD)/steatohepatitis (NASH) and atherosclerotic cardiovascular disease. Macrophages integrate metabolic and immune responses and thus represent key effector cells for these pathophysiologies.

In the present project, the impact of signal transduction pathways controlling central checkpoints of metabolism on NAFLD/NASH and atherosclerosis will be explored in experiments spanning from cell culture to in vivo models of disease (including bone marrow transplantation and gene transfer with recombinant viral vectors). CRISPR/Cas9 knockout screens and RNA sequencing will be carried out with the aim to discover and map novel signal transduction pathways. These approaches are complemented with lipid biochemistry and mass spectrometry determinations of changes in metabolites.

Diabetic foot ulcer (DFU) is a major complication of diabetes mellitus, and increased understanding of its molecular pathogenesis is needed to develop more effective treatments. Circular RNA (circRNA) is a class of RNA with the 3′ and 5′ ends covalently linked, which widespread in eukaryotes has only been recognized lately; however, increasing evidence has demonstrated their important roles in health and diseases. A novel type of mitochondria-encoded circRNAs (mecciRNAs) has been discovered in humans and mice recently. The purpose of this research project is to unravel the role(s) of mecciRNAs in skin wound healing and to explore the therapeutic potential of modulating circRNAs in DFU. The findings of this study will, from a new angle, add to our understanding of the pathogenesis of diabetic ulcers and may uncover novel drug targets for further clinical studies.

Postdoctoral position summary/title: Postdoctoral Researcher/The Strategic Research Programme for Diabetes.

Mitochondria are a central node for the integration of nutrient and energy homeostasis and are highly dynamic organelles that cope with oscillations in energy demand imposed by energetic stressors throughout the day. Preliminary results from our group indicate that mitochondrial rhythmicity of respiration is lost in skeletal muscle from type 2 diabetic patients. The mechanisms underlying disrupted circadian rhythmicity of the intrinsic molecular clock in type 2 diabetes are currently unknown. The main objective of this project is to determine whether there is bidirectional signaling between the core clock and pathways coordinating mitochondrial metabolism, and the extent to which the circadian transcriptional machinery is disrupted in type 2 diabetes. These questions are clinically relevant given the dynamic diurnal changes in a plethora of hormones, metabolites and substrates. The postdoctoral fellow will adopt multi-disciplinary approaches, integrating animal models and cell-based systems with clinical material from type 2 diabetic patients to identify and functionally validate candidate transcription factors and transcriptional pathways for intervention to restore circadian rhythmicity in type 2 diabetes. This project will also include usage of RNA-seq, ChIP-seq and interaction proteomics as well as respirometry and microscopy. The successful candidate will receive necessary basic training within these technologies.

The overarching goal of this translational project is to define the molecular fingerprints of reno-protective drugs at the single cell level in kidney tissue, with the purpose of identifying underlying cellular mechanisms that slow the progression of nephropathy. We use single cell RNA sequencing to extensively profile mouse/human kidney tissue and urine samples in healthy and diabetic conditions. Studies will be complemented with experiments in kidney organoids. Key molecular mechanisms identified will be validated and investigated further using transgenic mouse models.

Postdoctoral position summary/title: Postdoctoral scholarship: Translational asthma research on lipid mediator regulation of small airway inflammation.

The goal is to define mechanisms of relevance to asthma that ultimately may become novel targets for future treatments. This post doctoral fellow will be responsible for a project where specific interactions between lipid mediators and key cytokines in type 2 as well as non-type 2 inflammation will be investigated (for model, see e.g. Manson et al J Allergy Clin Immunol. 2020; 145:808-81) using isolated tissues from the small airways of humans and guinea pigs. This is a physiological and pharmacological research project, but includes supporting techniques such as histology, transcriptomics, proteomics, and the measurement of mediator release using different methods.

The Karolinska Institute sometimes known as the (Royal) Caroline Institute in English is a research-led medical university in Solna within the Stockholm urban area of Sweden. The Nobel Assembly at the Karolinska Institute awards the Nobel Prize in Physiology or Medicine. The assembly consists of fifty professors from various medical disciplines at the university. The current rector of Karolinska Institute is Ole Petter Ottersen, who took office in August 2017.

The Karolinska Institute was founded in 1810 on the island of Kungsholmen on the west side of Stockholm; the main campus was relocated decades later to Solna, just outside Stockholm. A second campus was established more recently in Flemingsberg, Huddinge, south of Stockholm.

The Karolinska Institute is Sweden’s third oldest medical school, after Uppsala University (founded in 1477) and Lund University (founded in 1666). It is one of Sweden’s largest centres for training and research, accounting for 30% of the medical training and more than 40% of all academic medical and life science research conducted in Sweden.

The Karolinska University Hospital, located in Solna and Huddinge, is associated with the university as a research and teaching hospital. Together they form an academic health science centre. While most of the medical programs are taught in Swedish, the bulk of the PhD projects are conducted in English. The institute’s name is a reference to the Caroleans.
