Diabetes features a reduction in the number of functional β-cells, a key pathologic event that causes or exacerbates the dysregulation of blood glucose levels. Regenerating insulin-producing β-cells might prove a better treatment for diabetes, which is at present controlled but not reversed by insulin injections. Therefore, the main interest in the lab of Olov Andersson is to determine how small molecules and cellular mechanisms can promote regeneration of insulin-producing beta-cells and improve metabolic control, as potential curative approaches for diabetes. The successful applicant will use chemical-genetic screening techniques in zebrafish to identify compounds, genes and signalling pathways that can promote β-cell regeneration.

During the development of obesity and insulin resistance, ectopic lipid accumulation occurs in various organs. In the liver, such condition is called non-alcoholic fatty liver disease (NAFLD), which can progress to more severe stages, such as non-alcoholic steatohepatitis (NASH), cirrhosis, and/or hepatocellular carcinoma. Liver fibrosis is the most important predictor of prognosis in NAFLD patients. In this project, we will focus on NAFLD and develop an innovative non-invasive machine learning model to monitor the real-time disease progression and future risks of NAFLD patients. The tools developed here will allow patient subgroup stratification at a high resolution and further investigate how the immune system is functional in high-risk NAFLD patients during their longitudinal disease progression.

Postdoc Fellowship Position summary/title: Postdoctoral position: Insulin nanotechnology (4)
At the Teixeira lab (Ana Teixeira-group), we have developed DNA nanotechnology-based approaches to map and target membrane protein nanodomains (e.g. Ambrosetti et al, Nat Nanotechnology, 2021; Fang et al, ACS Nano, 2021; Verheyen, Fang et al, Nucleic Acids Research, 2020; Shaw, Lundin, et al, Nat Methods 2014). In this project, the postdoctoral research fellow will investigate the nanoscale spatial organization of insulin receptors in different tissues. This information will be used to develop multivalent insulin variants using DNA- and protein-based self-assembly. The biodistribution and tissue specific effects of different multivalent insulin variants will be mapped in wild type and diabetes models in zebrafish and mice. This work aims to form the basis for the future development of insulin variants with tailored tissue targeting patterns.

Postdoc Fellowship Position summary/title: Postdoctoral studies in translational immunology focusing on autoimmunity and lymphoma in the skin (scholarship)
Our main research interest is to understand pathways governing development of systemic inflammatory diseases and lymphoma and the link between those two, since patients with autoimmune diseases run an increased risk of developing lymphoma and other cancers. We will investigate possible immune mechanisms and tumor properties governing cancer development in patients with autoimmunity, including dermatomyositis and systemic lupus erythematosus. In addition, we focus on cutaneous T cell lymphoma, which is a rare type of lymphoma primarily engaging the skin. The unique accessibility of this disease allows for easy access to the tumor and the possibility to follow the development of the lymphoma and anti-lymphoma immune responses over time. Most of the patients with cutaneous T-cell lymphoma get an indolent disease, but for largely unknown reasons around 10-15% percent develop aggressive disease with high fatality. In these efforts our ultimate aims are to increase fundamental knowledge on the pathogenesis of cutaneous lymphoma and cancer in autoimmunity and aid earlier identification of patients at risk of aggressive disease.

These targets will then be used for the development of a delivery system to target diseased cells in vivo. Most of the research will be conducted utilizing proteomic and sequencing-based techniques. The first part will involve isolating EVs from patient biofluids, from which therapeutic targets will be identified. Following validation of these targets, a drug delivery system utilizing lipid nanoparticles targeted to the previously identified extracellular proteins will be developed. Successful candidate constructs will later be developed to target disease relevant targets in vivo.

The Karolinska Institute sometimes known as the (Royal) Caroline Institute in English is a research-led medical university in Solna within the Stockholm urban area of Sweden. The Nobel Assembly at the Karolinska Institute awards the Nobel Prize in Physiology or Medicine. The assembly consists of fifty professors from various medical disciplines at the university. The current rector of Karolinska Institute is Ole Petter Ottersen, who took office in August 2017.
The Karolinska Institute was founded in 1810 on the island of Kungsholmen on the west side of Stockholm; the main campus was relocated decades later to Solna, just outside Stockholm. A second campus was established more recently in Flemingsberg, Huddinge, south of Stockholm.
The Karolinska Institute is Sweden’s third oldest medical school, after Uppsala University (founded in 1477) and Lund University (founded in 1666). It is one of Sweden’s largest centres for training and research, accounting for 30% of the medical training and more than 40% of all academic medical and life science research conducted in Sweden.
The Karolinska University Hospital, located in Solna and Huddinge, is associated with the university as a research and teaching hospital. Together they form an academic health science centre. While most of the medical programs are taught in Swedish, the bulk of the PhD projects are conducted in English. The institute’s name is a reference to the Caroleans.
