The postdoc will engage in a collaboration that aims to study the intra- and intergenerational health and labour market implications and costs of divorce among families in Denmark. The postdoc will develop novel statistical methods and apply these to register and RCT data to investigate heterogeneity in effects of divorces and investigate whether subgroups are especially adversely affected by divorce. Specifically, the project aims to develop methods for inference on key features of the heterogeneous effects, to estimate characteristics and differences of the most and least impacted groups, and to data-adaptively identify key drivers of treatment effect heterogeneity. In a series of experiments, we have consistently shown that tasks-sets are competing biased by their importance and that this selection process is severely capacity limited. We will extend our new experimental procedures and mathematical models to the study of selection in planning and problem-solving. If our conjecture is true, higher-order cognitive processes like task selection, planning, and problem-solving depend on the same type of selection.

The aim of the project is to use the analytical tools of philosophy of science to assess the claim that absence of proper theorizing is the root cause of low replicability and to understand the type of theory needed to alleviate the crisis.

Postdoc Fellowship Position summary/title: Postdoc in Bioinformatics: Structural variations associated to circular DNA in cancer
Extrachromosomal DNA circles are known from tumors where they carry proto-oncogenes and can induce tumorigenesis (DOI: 10.1016/j.tig.2022.02.007). Our recent data reveal that circular DNA is also found in blood plasma as circular circulating tumor DNA (circular ctDNA), suggesting that this new type of ctDNA can be an efficient marker for cancer. We now aim to find patterns of the circular DNA in blood plasma and tumors from our thousands of samples that can serve as markers for cancer, relapse and prognosis of cancer. We will put emphasis on applying and developing cutting-edge bioinformatics methods for identification of patterns in circular DNA and novel structural circular DNA variants. The development of the methods for will include clustering and machine learning methods with output allowing for biological interpretations of the roles of circular DNA in cancer.

The position is funded by a Villum Young Investigator research grant and a Revive&Restore/Morris Animal Foundation grant to Assistant Professor Josefin Stiller. The postdoc will work on two projects. The first project involves a large-scale genomic dataset of hundreds of whole genomes of Syngnathiformes that our lab is generation, on which the postdoc will perform phylogenomic analyses to understand the evolutionary relationships and the sources of phylogenetic incongruence. These phylogenetic insights will be important for ongoing comparative genomics analyses on this dataset. Secondly, the postdoc will lead a conservation genomic project that aims to investigate connectivity, adaptive potential, and genetic diversity of leafy and common seadragons using whole genome resequencing for ca. 200 individuals. The growing obesity epidemic has escalated the progression of a series of non-communicable diseases, including type 2 diabetes and non-alcoholic steatohepatitis (NASH). While efficient pharmacotherapies have been developed for management of type 2 diabetes, there are no medications approved to treat NASH or for its predecessor non-alcoholic fatty liver disease (NAFLD). This project is based on an interdisciplinary collaborative approach between the Clemmensen Group and the Sakamoto Group and includes medicinal chemistry, in vitro/cellular pharmacology, metabolic signaling and preclinical pharmacology. We have developed first-generation liver-targeting pharmacological candidate molecules for safely targeting hepatic metabolic processes to improve liver health in context of fatty liver diseases (NAFLD and NASH). We have preliminary proof of principle in vitro and in vivo data and we are now eagerly looking to accelerate further preclinical testing in appropriate genetic and disease models relevant to liver and other metabolic diseases, including loss-of-function models to confirm target engagement in vivo. As such the project bridges from basic understanding of molecular mechanisms to translation and innovation. Your key tasks will be to design and perform in-depth in vivo pharmacology and metabolic/biochemical analysis of various strains/disease conditions of animals treated with the drug of our interest.

The main objectives of this post-doctoral project are to elucidate the interplay between energy sensing pathways (predominantly AMPK) and the circadian clock to regulate systemic metabolism. To understand the impact of AMPK on circadian biology, skeletal muscle-specific AMPKγ3 knockout and AMPKγ3 transgenic mice, and respective wild-type mice, will be studied. Mice will be studied across a 12h:12h light:dark cycle to characterize the role of AMPK in regulating circadian metabolism. The role of AMPK in regulating mitochondrial rhythmicity will be investigated by crossing mito-QC mice with AMPKγ3 knockout and transgenic mice, and studying mitochondrial respiration, mitophagy, and mitochondrial content across the circadian cycle. Finally, we will determine the role of skeletal muscle AMPK in the circadian response to energetic stressors via timed exercise and fasting in AMPKγ3 knockout and transgenic mice.

A major challenge in academia and in the pharmaceutical industry is to transform early-stage projects into valuable assets through effective target validation studies. As a postdoctoral researcher, you will be the expert in validating the physiological role and pharmacological potential of GPR39 in prevention and treatment of metabolic disorders. To achieve this, you will utilize agonist leads developed in-house in ex vivo and in vivo target validation studies. The prototype agonist leads have already been scaled up in amounts suitable for long term chronic in vivo studies in rodents, enabling a series of unique target validations studies of increasing complexity. To ensure that your research has maximum impact, you will establish collaborations with other research groups at the Center, enabling you to take full advantage of the state-of-the-art technologies available, especially at the Rodent Metabolic Phenotyping Platform. Your primary responsibilities will involve characterizing the mechanism of action of the agonist leads and build a strong efficacy package, with the ultimate goal of validating the physiological role of GPR39 in regulating, type-2 diabetes, NAFLD and ALD.
