(1) The tumor suppressor p53, metabolism, and autophagy. We are interested in the regulation and functions of the preeminent tumor suppressor p53. Our results have revealed a role for p53 in modulating metabolic pathways that are critical for biosynthesis and redox balance. We are investigating the function of p53 as both a sentinel and a regulator for metabolic activities. We are also identifying and characterizing metabolic alterations that drive tumor initiation and progression. A recent extension of this research area is to define the role of metabolism and autophagy in stem cells, including embryonic stem cells and cancer stem cells.
(2) Protein quality control, aging, and neurodegeneration. Our lab recently identified two protein quality control (PQC) systems, which consist of tripartite motif (TRIM) proteins and poly-Asp/Glu (polyD/E) proteins, respectively. Unlike canonical PQC systems, these new systems are independent of ATP and are unique for animals (the TRIM system) or eukaryotes (the polyD/E system). Both systems are multifunctional and highly effective. We are investigating their mechanisms of action; their roles in aging and neurodegenerative diseases, including Alzheimer’s disease, Parkinson’s disease, and amyotrophic lateral sclerosis; and their utility in treating these diseases.

The Nathanson laboratory invites applications for Post-Doctoral positions at the Perelman School of Medicine, University of Pennsylvania. We focus on cutting-edge translational research in cancer genetics, both in the inherited and somatic genetics of cancer (example publications include Hu et al., NEJM 2021; Maxwell et al., JCO Pres Onc, 2020; Kraya et al., Clin Cancer Res 2019). Expertise and interest in both wet bench studies and bioinformatics is preferred, however applicants with only expertise in bioinformatics would be welcome. The laboratory is highly collaborative and investigates several cancers, among which are hereditary breast cancer (due to BRCA1/2 and non-BRCA1/2 mutations), melanoma, testicular germ cell tumors, and neuroendocrine tumors. Applications for postdoctoral positions are invited across all areas of research, all of which are funded by multiple NIH grants and foundation funding. Potential projects include research to identify novel susceptibility variants through whole exome sequencing in men with testicular germ cell tumors, on the inherited and somatic genetics of breast cancer, including identification of novel susceptibility genes, characterization of moderate penetrance genes, and somatic studies of breast and other cancers in BRCA1/2 mutation carriers (with bulk and single cell RNA/DNA sequencing, and immunophenotyping), with the opportunity to perform correlative studies in relationship to therapeutic response, both targeted and immunotherapies. Further, we have access to the Penn Medicine Biobank with over 45K individuals with whole exome sequencing available for multiple genetic studies in cancer. The laboratory is located with the Institute for Immunology at Penn, and there are multiple opportunities for local collaboration. Applicants also will have the opportunity to engage in collaborative efforts at the national and international stage.

A post-doctoral position in human genetics and genomics is immediately available. Research in our group focuses on the development of genomics and bioinformatics methods to analyze sequencing data, and to enable the implementation of genomic medicine in clinical settings. The successful candidate will work on developing novel genomic methods to study human diseases that are related to genomic repeats. Example projects include the use of Illumina sequencing, optical mapping or long-read sequencing to assay human genomes, transcriptomes and epigenomes from patients with undiagnosed diseases, and to understand how repetitive elements and structural variants in complex genomic regions cause human diseases. Dr. Chesi has a long-standing interest in genomics and neurodegenerative diseases; she has developed Capture C based approaches to map GWAS signals to their effector genes (see some examples in linked publications) and is applying them to brain-related disorders.
Main areas of research in our lab are 3D genomics and epigenomics techniques (such as Hi-C, ATAC-seq, ChIP-seq and RNA-seq), CRISPR/Cas9 editing, pooled CRISPRi/a screens with scRNA-seq readout, plus development of iPSC-based models for neurodegenerative and neurodevelopmental diseases (2D neuronal/glial differentiation, co-cultures, 3D brain organoids).
