Postdoc Fellowship Position summary/title:  Post Doctoral Scholar

Zihai Li, M.D., Ph.D. – founding director of the Pelotonia Institute for Immuno-Oncology (PIIO) – is recruiting multiple postdoctoral scholars to join his research team at The Ohio State University Comprehensive Cancer Center (OSUCCC). Dr. Li’s research interests over the last 20-plus years have been primarily in the fields of chaperone biology, immune tolerance, and cancer immunology. The Li lab currently focuses on improving existing immuno-therapeutics and developing novel treatment strategies by understanding how the immune system influences cancer progression and the tumor microenvironment (https://go.osu.edu/zihai).  Supported by multiple grants from the NIH and private foundations, active projects include: elucidating the immune-mediated mechanisms behind sex biases in cancer; developing TGFβ-targeted therapies for cancer using anti-GARP monoclonal antibodies, CAR-T, and CAR-NK cells; studying the roles of mitochondrial metabolism in mediating response to radiation and immunotherapy to overcome resistance to PD-1 immune checkpoint blockers; uncovering immunoregulatory roles of platelets and other understudied myeloid cells in immune evasion; and understanding the fundamental mechanisms of differentiation program of regulatory T cells and CD8+ T cells in cancer, among others. Candidates are encouraged to pursue independent interests within the general framework of immuno-oncology, under the mentorship of Dr. Li. Members of the Li lab research team have opportunities for collaborations within the areas of emphasis in the PIIO Centers for Research Excellence including, Cell Therapy, Immuno-Genomics, Systems Immuno-Oncology and Translation Immuno-Oncology.

We are looking for skilled and highly motivated postdoctoral scholars with expertise in biochemistry, cell biology, structural biology, biophysics or related areas to conduct cutting-edge research on several projects focused on the role of actin binding proteins (e.g., plastins) and actin-specific bacterial toxins in the pathogenesis of human hereditary and infectious diseases, and cancer. Two examples of the lab’s priority projects are the recent discoveries of actin processive polymerization at the pointed end by Vibrio toxins (Sci. Adv., 2022) and the allosteric regulation of actin bundling proteins plastins (NSMB, 2022). Both, plastin and toxin projects relate to the mechanotransduction/mechanosensing properties of the actin cytoskeleton. Dr. Vladi Karniychuk’s laboratory at the Department of Veterinary Biosciences, is seeking a highly-motivated and dedicated to the profession postdoctoral scholar to conduct scientific research in virology. Increasing CpG dinucleotide numbers in RNA viral genomes, while preserving the original amino acid composition of encoded proteins, is a cutting-edge approach for live vaccines. CpG-recoded viruses evoke impaired infection which does not cause disease. Beneficially, this impaired infection leads to robust antiviral host immunity. Our lab developed several CpG-enriched Zika virus vaccine candidates and demonstrated safety and protective efficacy in neonatal and adult mouse models and during mouse pregnancy [Trus et al. Methods Mol Biol, 2022; Udenze et al. Frontiers in Immunology, 2022; Trus et al. Frontiers in Immunology, 2020]. In addition to vaccine studies, we were the first to demonstrate the oncolytic potential of CpG-enriched RNA viruses. Specifically, we showed that Zika virus variants with increased genomic CpG content have reduced infection in healthy brain cells but high virulence in glioblastoma stem cells—stem cells that can self-renew and initiate the growth of glioblastoma, the most aggressive type of cancers [Trus et al., Viruses, 2020]. A postdoctoral fellow on the current project will study how the CpG dinucleotide content in viral RNA affects interactions with cellular proteins and immune responses. This will help to develop a better CpG enrichment approach for vaccines and oncolytics, and contribute new fundamental knowledge on viral genome biology. The project employs a range of molecular biological and virology approaches, cell culture, immunoprecipitation, and proteomics. A particularly ambitious and productive postdoctoral scholar will also have an opportunity to contribute to our flavivirus pathogenesis studies. The Ohio State University, commonly Ohio State or OSU, is a public land-grant research university in Columbus, Ohio. The flagship of the University System of Ohio, it is considered a Public Ivy, and has been ranked by major institutional rankings as among the best public universities in the United States. Founded in 1870 as the state’s land-grant university and the ninth university in Ohio with the Morrill Act of 1862, Ohio State was originally known as the Ohio Agricultural and Mechanical College and focused on various agricultural and mechanical disciplines but it developed into a comprehensive university under the direction of then-Governor and later U.S. president Rutherford B. Hayes, and in 1878 the Ohio General Assembly passed a law changing the name to “the Ohio State University” and broadening the scope of the university. Admission standards greatly tightened and became more selective throughout the 1990s and 2000s.

Ohio State’s political science department and faculty have greatly contributed to the construction and development of the constructivist and realist schools of international relations; a 2004 LSE study ranked the program as 1st among a public institution and 4th overall in the world. A member of the Association of American Universities, Ohio State is a leading producer of Fulbright Scholars, and is the only school in North America that offers an ABET-accredited undergraduate degree in welding engineering. The university’s endowment of $5.29 billion in 2020 is among the most in the world. Past and present alumni and faculty include 5 Nobel Prize laureates, 9 Rhodes Scholars, 7 Churchill Scholars, 1 Fields Medalist, 7 Pulitzer Prize winners, 64 Goldwater scholars, 6 U.S. Senators, 15 U.S. Representatives, and 104 Olympic medalists. It is classified among “R1: Doctoral Universities – Very high research activity”. As of 2021, Ohio State has the most number of students in the United States who are in the 95th percentile or above on standardized testing of any public university.
