Postdoc Fellowship Position summary/title:  Translational Neurosurgical Oncology Postdoctoral Researcher

The focus of our lab’s research is at the intersection of immunology, metabolism, and brain tumor biology. The primary goal of our translational research is to understand mechanisms of immune evasion by primary malignant brain tumors and skull base tumors to generate novel immune-based therapies. We utilize a combination of high-dimensional single cell immune and genomic profiling, metabolomics, and advanced tumor organoid and mouse models, to 1)
understand the role of myeloid derived suppressor cells in promoting tumor growth and T cell dysfunction, 2) elucidate the mechanisms of tumor-specific T cell exhaustion and clonal lineage of dysfunction T cells, and 3) understand how metabolic networks control immune cell function within the tumor microenvironment. Finally, our lab is uniquely integrated with the Penn Brain Tumor Center, as such, our lab is involved in evaluating immune responses in brain tumor patients undergoing immunotherapy with the goal of identifying predictors of response and new immune targets. Primary responsibilities include leading the implementation of experimental projects, responsible for the design of the translational/basic science experiments to test hypotheses. The candidate will have a leadership role and will be responsible for the supervision of productivity of other laboratory members, provide training as necessary, and also collaborate with other groups on campus. The candidate will be independent, self-driven, organized, and possess excellent managerial and scientific writing skills. The highly qualified candidate will have excellent opportunities for growth, independence, and teaching in a dynamic lab with the goal to transition to independence. (02) Postdoctoral Fellowship Position

Postdoc summary/title:  Postdoctoral Researcher Positions in Cancer Biology

Our research is dedicated to unraveling the contribution of recurrent genomic alterations, both in protein-coding and non-coding regions, to tumorigenesis. Our primary focus is on identifying therapeutic targets and developing combination strategies for the treatment of cancer. To achieve these goals, we have developed a novel computational approach that integrates large-scale genomic, functionomic, and pharmacologic profiles. This innovative methodology has led to the creation of the Functional Cancer Genome data portal (FCG, http://fcgportal.org/home/), which serves as a comprehensive resource for functionally annotating cancer genomes. The newly funded projects will build upon the novel discoveries generated by the FCG project and will involve in-depth molecular characterization of new epigenetic and immuno-oncology drivers in cancer. We are currently recruiting for three related but independent projects: 1) Epigenetic dysregulation and non-coding RNAs in tumorigenesis, 2) Tumor immunology and immune therapy, and 3) Targeted and epigenetic therapy for cancer treatment. To conduct these projects, we employ cutting-edge technologies such as single-cell and spatial RNA sequencing analysis, as well as CRISPR screen and high-throughput chemical screen technologies. Additionally, our lab’s computational biology team provides a wealth of novel computational biology approaches and resources to support these research projects.

We are a multi-disciplinary and collaborative research group seeking highly-motivated investigators to join our program studying rare genetic human diseases of extra-skeletal (heterotopic) bone formation. Inactivating mutations in the human GNAS gene cause POH; heterotopic bone in POH initiates within subcutaneous adipose tissue and is preceded by depletion of the adipose tissue and lipid content. Activating mutations in ACVR1, a BMP type I receptor, cause FOP; in skeletal muscle heterotopic ossification is preceded by an altered immune response and changes in the tissue microenvironment. A Postdoctoral Researcher position is available for a highly motivated applicant in the laboratory of Dr. Fiona Simpkins at the University of Pennsylvania. Our focuses are to understand resistance mechanisms to targeted therapies such as DNA Damage Repair inhibitors (DDRi), to sequence DDRi with Immuno-Oncology (IO) drugs, and to identify biomarkers or response to these agents. We have developed a preclinical drug development program in the Ovarian Cancer Research Center with the goal of moving novel laboratory therapeutic discoveries into the clinic. We have developed >150 patient derived xenografts (PDX) models, patient-derived organoids, and primary tumor cultures from ovarian and endometrial cancers. These PDX models mimic the original tumors, thereby accelerating the translation of preclinical study results into clinical trials for patients. Also, we are “humanizing” our models by autologous tumor infiltrating lymphocyte (TIL) and macrophage (TAM) transfer, creating an autologous in vivo model system. Our laboratory has close collaborations with pharmaceutical companies including AstraZeneca, Merck, REPAIR, Zentalis. Based on preclinical studies from our lab, novel combination therapies have moved to phase 1/2 clinical trials led by Dr Simpkins (e.g. ClinicalTrials.gov Identifier: NCT03462342 and GY031). We have another two drug combinations also moving into clinical trials. Dr. Simpkins strongly supports career development of postdocs, either developing an independent laboratory in academia or finding a position in industry. We provide a rich multi-disciplinary training environment in animal modeling, molecular biology, genomics, cancer biology, and immunology, and we have sufficient fundings from various sources, including NIH funding to support this. Previous trainees have been awarded with multiple grants including NIH career development K12 grant, OCRA early career grant, AACR fellowship, Rivkin award, OCRA mentored award, FWC grant, KOH grant and Harrison award.

The Volk laboratory invites applications for a postdoctoral position in our comparative breast cancer program at the School of Veterinary Medicine, University of Pennsylvania. The goals of our laboratory are to understand regulatory mechanisms governing dynamic interactions between cells and their surrounding extracellular matrix in the wound healing-fibrosis-cancer progression triad and to apply this knowledge to develop innovative regenerative and oncologic therapies for veterinary and human patients. One-year positions are available immediately with the option to extend an additional 1-3 years. Using a combination of in vitro and mouse models, as well as human, feline and canine breast cancer biopsy samples, we have initiated multiple lines of investigation that will provide insight into how regulation of the collagen fibrillar matrix and its components, specifically type III collagen, inhibits formation of a tumor-permissive microenvironment. This project capitalizes on our studies using spontaneous naturally occurring large animal models of mammary tumors. Future work will characterize drivers of tumor-permissive and suppressive collagen signatures and investigate the use of a novel tissue engineering strategy to control tumor growth and metastasis following tumor resection to improve clinical outcome in veterinary and human breast cancer patients.

The Beers’ Lab has an existing program funded by NIH, the Department of Veterans Affairs, private foundations, and industry to study the molecular mechanisms that regulate tissue injury and repair in focused on discovery, target ID, target validation, and proof or principal studies aimed at uncovering and exploiting novel pathways to treat fibrotic lung diseases. In addition to the unique mouse reagents, the successful candidate will have access to a variety of cutting edge technologies including RNAseq, single cell and ATAC RNAseq, high resolution proteomics, CRISPR mediated screens, lineage tracing, organoid assays, and iPS cells to identify pathways critical for lung tissue repair and regeneration. The ideal candidate will have an M.D., Ph.D. or MD/PhD with a strong background in basic molecular and cell biology skills. Experience utilizing mouse models and primary cell culture systems is required for the success of this position. The PPCLBI is an interdisciplinary center that combines basic, clinical and translational research being conducted by Pulmonary faculty, both at UPenn (adult patients) and at Children’s Hospital of Philadelphia (neonatal and pediatric patients). The LBI is housed in a completely renovated, state of the art research space in Stemmler Hall with more than 12,000 square feet of newly designed open concept lab space, offices and support areas. In addition to the research lab space, the PLBI houses the Human Lung Biology Core (HLB), which provides support in the design and implementation of experiments involving the use of human lung samples. The HLB Core has developed optimized protocols for the isolation and generation of high quality, reproducible human primary cells and organoids. The PCLBI houses eight core research labs with a wide range of basic and translational research including developmental biology, tissue regeneration, oncology, immunology, and inflammation.
