(02) Postdoctoral Fellowship Position

Postdoc summary/title: Postdoctoral Researcher/The Strategic Research Programme for Diabetes (5) Spatially resolving the response to anti-hyperglycemic drugs in adipose tissue

Increased white adipose tissue mass is linked to type 2 diabetes, which in turn can be treated with several different anti-hyperglycemic drugs. However, there are large interindividual variations in the efficacy and side-effects of these interventions. We hypothesize that distinct adipocyte subtypes determine insulin sensitivity and outcomes following interventions with anti-hyperglycemic drugs. This project will test our model by spatially resolving treatment responses in human WAT based on samples from a clinical trial called DIASPAX (NCT05501483). In the study, we obtain adipose tissue biopsies, which we will use to generate single-nucleus RNA sequencing and spatial transcriptomic data. By jointly analyzing these data together with functional assessment of the tissue and assessments of clinical phenotypes, we will for the first-time link white adipose tissue heterogeneity to treatment responses. We are now looking for an ambitious postdoctoral fellow that will perform single-nucleus RNA sequencing and spatial transcriptomic analyses. The work will be undertaken in collaboration with other experts within the group as well as with members of the lab of Patrik Ståhl at SciLifeLab. There will be possibilities to follow-up key findings with wet lab experiments using state-of-the-art methods.

(05) Postdoctoral Fellowship Position

Postdoc Fellowship Position summary/title: Postdoctoral Researcher/The Strategic Research Programme for Diabetes (2) Local and systemic impact of adipose tissue on vascular function in cardiometabolic disease

People living with type 2 diabetes are twice as likely to develop heart disease or stroke than those without diabetes, yet the precise mechanisms for this increased atherosclerosis risk remain poorly understood. This translational postdoc project aims to shed light on the links between diabetes, obesity and the associated heightened risk for atherosclerosis by exploring the differential effect of local adipose tissue surrounding larger vessels (perivascular fat) and the systemic impact of the subcutaneous adipose tissue, which represents around 80% of the human fat mass. The project will explore both known disease pathways such as Arginase activity and identify novel mediating factors affecting vascular function by collecting paired clinical fat biopsies and performing molecular and functional analyses. The results can potentially have large implications for our understanding of the interconnection between diabetes and cardiovascular risk, and help identify valuable therapeutic targets for future protective treatments for this extra vulnerable patient group.

(07) Postdoctoral Fellowship Position

Postdoc Fellowship Position summary/title: Postdoctoral Researcher/The Strategic Research Programme for Diabetes (4) Dietary nitrate and physical exercise to prevent cardiovascular disease in diabetes

Physical exercise and dietary improvements are key non-pharmacological strategies for metabolic syndrome and diabetes. These lifestyle changes reduce chronic oxidative stress and increase nitric oxide (NO) bioavailability. Our research focuses on inorganic nitrate, found in green leafy vegetables and beetroot, which converts to bioactive NO. Studies show nitrate improves cardiovascular function, reducing blood pressure, improving vascular function, and enhancing exercise parameters like mitochondrial efficiency and oxygen utilization. We hypothesize that dietary inorganic nitrate combined with physical exercise will provide significant cardiovascular protection for T2D patients, who are at high risk for CVD. This clinical trial will study the combined effects of structured physical exercise and dietary nitrate supplementation on cardiovascular health in T2D patients.

(09) Postdoctoral Fellowship Position

Postdoc Fellowship Position summary/title: Postdoctoral Researcher/The Strategic Research Programme for Diabetes (6) Signal transducers regulating glucose transport in type 2 diabetes

The Integrative Physiology research group at the Department of Molecular Medicine and Surgery at Karolinska Institutet is seeking a highly motivated postdoctoral researcher to join the group of Professor Juleen R. Zierath. The research in the group is focused on translational studies on cellular and molecular mechanisms underlying insulin resistance and type 2 diabetes and the effects of physical activity on glucose and lipid metabolism in skeletal muscle.

This project is designed to test the role of signal transducers to improve glucose homeostasis by targeting skeletal muscle glucose transport. The candidate will explore the molecular signal transducers involved in the regulation of glucose transport to restore glucose uptake in skeletal muscle from people with type 2 diabetes. The team has identified several candidates that may play a direct role in glucose transport. The postdoctoral fellow will validate the role of novel signal transducers in the regulation of glucose transport and insulin sensitivity using knock-down and over-expression approaches in primary human skeletal muscle cells. Promising candidates will be further validated in in vivo models of type 2 diabetes through electroporation approaches.

About The Karolinska Institute, Sweden – Official Website

The Karolinska Institute sometimes known as the (Royal) Caroline Institute in English is a research-led medical university in Solna within the Stockholm urban area of Sweden. The Nobel Assembly at the Karolinska Institute awards the Nobel Prize in Physiology or Medicine. The assembly consists of fifty professors from various medical disciplines at the university. The current rector of Karolinska Institute is Ole Petter Ottersen, who took office in August 2017.

The Karolinska Institute was founded in 1810 on the island of Kungsholmen on the west side of Stockholm; the main campus was relocated decades later to Solna, just outside Stockholm. A second campus was established more recently in Flemingsberg, Huddinge, south of Stockholm.

The Karolinska Institute is Sweden’s third oldest medical school, after Uppsala University (founded in 1477) and Lund University (founded in 1666). It is one of Sweden’s largest centres for training and research, accounting for 30% of the medical training and more than 40% of all academic medical and life science research conducted in Sweden.

The Karolinska University Hospital, located in Solna and Huddinge, is associated with the university as a research and teaching hospital. Together they form an academic health science centre. While most of the medical programs are taught in Swedish, the bulk of the PhD projects are conducted in English. The institute’s name is a reference to the Caroleans.
