We are seeking a talented and enthusiastic Research Assistant / Research Associate to join the CAPKR at the University of Manchester. The successful candidate will become a member of the CAPKR research team and be fully involved in a project entitled “Defining Local Disposition Beyond What is Known for Small Molecules to Assist Building PBPK Framework for Oligonucleotide and Antibody Drug Conjugate Modalities”. Small molecules have dominated drug therapy for over a century, but recent decades have seen the development of increasingly larger and more sophisticated drug types. These include both protein or peptide based drugs, such as antibodies, and oligonucleotides. The distribution of such molecules within cells, such as liver cells, is not well-understood. Oligonucleotides, for example, are known to distribute in lysosomes, cytoplasm and nucleolus.

We are seeking an enthusiastic and highly motivated Bioinformatician to collaborate with scientists and clinicians to provide knowledge and expertise in the analysis and interpretation of data from translational radiotherapy-related research. One of the goals of the MCRC Radiotherapy Related Research (RRR) group is to personalise radiotherapy (field, dose, fractionation, use of chemotherapy) by integrating tumour, patient and treatment information to optimise radiotherapy delivery for individual patients and improve outcomes over conventional treatments that approach all patients in a similar manner. A significant proportion of the work will focus on developing an improved understanding of the molecular basis of patient response to radiotherapy, and on decoding the human transcriptomes/ genomes/ proteins associated with the response of normal and malignant cells to radiotherapy.

(03) Postdoctoral Fellowship Position
Postdoc Fellowship Position summary/title: Research Associate – Pharmacoepidemiology and Causal Inference
Immune-mediated inflammatory diseases (IMIDs) include conditions affecting the skin, joints, and/or the gut. They lead to a big burden on the body and mind for 7% of people globally. New high-cost treatments that target inflammation-controlling proteins have transformed the lives of people with IMIDs. When deciding whether to take these treatments, patients and their doctors rely on evidence. This includes information from randomized controlled trials (RCTs), where two or more different medicines, of which one may be a dummy medicine, are given randomly to groups of participants. There are also observational studies, where large groups of people are observed in routine settings without being allocated medicines. Observational studies are representative, large, and relatively cheap but can be biased. RCTs are more reliable but people from underserved populations, such as people from lower socio-economic backgrounds, are often excluded or under-represented. Only using evidence from RCTs to guide treatment decisions could introduce privileges or disadvantages for different groups of people, leading to an overestimation of benefits and an underestimation of risks overall.

This project is led by Professor Sophie Nixon at the University of Manchester, in collaboration with the Earlham Institute. In this project we are interrogating the rules that govern microbiomes that inhabit terrestrial hot springs.
The long-held tradition of studying microorganisms in isolation has, until recently, obscured the fact that microbial life exists as interdependent multi-species assemblages. Despite the profound impact microbial communities have on their environment and the technological advances in microbial genomics in recent years, the rules that govern the structure, function and stability of microbial communities are complex and only partially understood. Although a plethora of mechanisms that influence microbial communities have been identified, from metabolic priming, to inter- and intraspecific metabolic signalling and even direct antibiosis, holistic understanding has yet to be achieved in any microbial system. In this project we will develop a novel experimental system that is complex yet tractable, enabling rich, coordinated multi-omics analysis coupled with computational modelling to deliver deep mechanistic insight. We will apply a cutting-edge toolbox of experimental, analytical, and computational approaches to reveal the rules of life in this system that will serve as a blueprint to apply to any microbiome.

Postdoc Fellowship Position summary/title: Research Associate Bioinformatician
We are seeking a talented and ambitious Bioinformatician to join the Wedge lab, which has a focus on Cancer Evolution and Heterogeneity. One of the key goals of this lab is to improve clinical practice by advancing our understanding of the processes and mechanisms involved in tumour progression. Using pioneering computational methods of DNA sequencing, the Wedge lab aims to generate genomic profiles of sub-populations within each tumour in order to track the evolutionary trajectories within tumours. This post will take a leading role in the analysis of data from high-throughput sequencing experiments, including Whole Genome Sequencing, in head and neck cancer during and after radiotherapy. The research involves the analysis of data from cancer patients and clinical samples. The analysis will form a part of a project that collaborates with researchers at the University of Cambridge and is funded by the CRUK RadNet programme.

Postdoc Fellowship Position summary/title: Research Associate in Generative AI for Biological Systems
Applicants are invited for the post of Postgraduate Research Associate in Generative Artificial Intelligence (AI) for Biological Systems. Generative models have been successfully applied across a broad range of biological systems. Recent applications have included modelling single-cell gene expression, biological sequences (protein, DNA and RNA), and microscopy images/movies.
The post-holder will develop probabilistic machine learning approaches for one or more of these data modalities. In the context of biology there are several challenges to applying generative models, e.g. uncovering causal relationships, incorporating prior knowledge, dealing with experimental variability, and fine-tuning of pre-trained models for specific applications. In this project we are particularly interested in using models to make decisions, e.g. to design further experiments, in which case it is important to associate model-based inferences with well-calibrated uncertainties.
