(04) Postdoctoral Fellowship Position

Postdoc Fellowship Position summary/title: Postdoctoral Research Associate in the Cellular Structural Biology of the Meiotic Synaptonemal complex

This position will study the cellular structural biology of the human synaptonemal complex. This position is ideal for a cell biologist with an interest in the structure of intra-cellular supramolecular protein assemblies. It will involve cellular biology (mammalian cell lines), widefield imaging, SIM/STORM imaging, TEM, cryo-electron tomography, sub-tomogram averaging, purification from cellular systems, and structural analysis (including cryo-EM) of purified complexes. Cryo-EM data will be collected at national facilities, including Diamond Light Source and the Scottish Centre for Macromolecular Imaging. The successful candidate will lead this project (including several PhD students), and will collaborate with researchers in the lab focussed on the structural biology of recombinant protein assemblies, to achieve our wider goals of understanding the integrated structure and function of the full mammalian synaptonemal complex. The successful candidate will be fully supported in applying for postdoctoral research fellowships within the lab, and independent research positions at the end of the postdoc. In suitable cases, part of the project, or a new project to be started during the employment, could be taken by the successful candidate as the basis for their future research career. Applicants should submit a document explaining their research interests, how these apply to the proposed project on the synaptonemal complex, and the vision for their future career.

Quantitative studies suggest that patients with severe mental illness may be less likely to be prescribed secondary prevention cardiovascular medications. However, there is little qualitative data in this area, and a lack of understanding of the challenges and/or barriers faced by GPs when treating this vulnerable group of patients. The post holder will work on a qualitative project grant aimed at gaining insight into the experiences of GPs, when treating people with SMI for CVD, to help us understand why people with SMI may be less likely to receive appropriate CVD medication. As well as informing planned larger mixed-methods studies in this area, findings will inform how best to support GPs to ensure they are delivering the best possible care, and most appropriate treatment, for people with both SMI and CVD. Key roles include participant recruitment, data collection (telephone or online on-to-one interviews), analysis of interview data and writing-up of findings for publication in peer-reviewed journals.

We seek an outstanding candidate to join the group of Dr. Tom Deegan, within a programme aimed at understanding the molecular mechanisms and regulation of eukaryotic DNA replication. We have developed biochemically reconstituted systems to interrogate the ill-defined termination phase of DNA replication at the molecular level (Deegan et al, Mol. Cell., 2019; Deegan et al, eLife, 2020), and have recently described a mechanism for eukaryotic replisome disassembly (Jenkyn-Bedford et al., Nature, 2021). A number of projects are now available, using biochemical reconstitution with purified yeast and human proteins, cryo-EM, and mammalian cell models to investigate molecular mechanisms of DNA replication termination, and how this process is subverted under the types of DNA replication stress that are characteristic of human cancer. Our group is part of the MRC Human Genetics Unit, a world class centre, which provides a highly collaborative and multi-disciplinary environment within which to study eukaryotic chromosome biology.

Prokaryotic growth can be modulated in different ways, including temperature, antibiotics, toxins, nutrients, and osmolarity. While the effects of nutrients have been extensively characterized, a common modulation of bacterial growth occurs in response to changes in external osmolarity, and it remains poorly understood. We hypothesize that the osmotically induced growth slowdown can be explained by examining several physiological variables and the successful applicant will measure them using both single-cell microscopy and microbiology population assays. She/he will integrate the results with a mathematical model as well. The post holder will also have the opportunity to investigate long term survival of bacterial cells at high osmolarities.

Reporting to, and working with, Dr Miguel O. Bernabeu you will conduct research centred around one of the two following areas: a) the develop of novel approaches to phenotyping retinal blood vessels [1] in animal models of stroke and investigating associations with brain phenotypes [2] and stroke outcomes, and b) the application mathematical models of vascular remodelling previously developed by the group [3, 4] to characterise the relationship between blood flow and endothelial cell migration in the development of arteriovenous malformations [5].
