PhD position summary/title: STI – 2 PhD positions in Interfacial Imaging of Water: New Light on Cellular Hydration
Nonlinear optical imaging and new ultrafast spectroscopic techniques have recently been developed in the Roke lab and used to image in real time interfacial water and electrostatic potentials on membrane interfaces of model membranes and in living cells.
In the current project: two new microscopes are to be constructed that will allow us to (1) image interfacial water in real time as well as the electrostatic field lines on the interface, and (2) image in real time membrane tension in vitro and in living systems. We will also provide a molecular ruler that links molecular conformation of lipids to mechanical forces. The new technology will first be applied in a controlled manner to in-vitro assays, after which measurements on living cells will be performed.

PhD position summary/title: STI – Two PhD student positions in Interfacial Imaging of Water: New Light on Cellular Hydration
Water is the liquid of life. It is intimately linked to our well-being. Without water, cell membranes cannot function. Charges and charged groups cannot be dissolved, self-assembly cannot occur, and proteins cannot fold. That water is intimately linked with life, we experience time and again when we quench our thirst, but how does this link work?
Osmosis is the flow of water across a (cell) membrane that separates two aqueous solutions with different concentrations of a solutes. Regulating osmotic pressure is a key survival strategy of cells and plays an important role in the functioning of every organism. How osmotic pressure and cell membrane tension are regulated on the molecular level is not known. It is the aim of the ERC Synergy Grant R2-Tension, a collaboration between EPFL (Prof. S. Roke) and the University of Geneva (Prof. A. Roux) to work this out. To do so, we will develop new technology and perform experiments in vitro and vivo.

The Polymers Laboratory at EPFL (lp.epfl.ch) has two open PhD positions for a project that focuses on the design and synthesis of block copolymers that will mimic the structure and properties of mussel-derived proteins.

The extension and transformation of our built environment are today responsible for a major part of greenhouse gas emissions, waste generation, and raw material extraction. While current climate crises call for reevaluating well-established dynamics in the construction and real-estate industries, circular economy strategies offer new opportunities to minimize the detrimental environmental impacts of buildings. But to what extent can they ensure the well-being of everyone within planetary boundaries?
Combining life-cycle assessment and urban twining methods with stakeholders interactions and expert knowledge on urban constructions and their long-term maintenance, the PhD thesis will (1) develop scenarios of change that public, commercial, and personal decision-makers could follow at the neighborhood, building, and dwelling scales in the short and long term horizons; (2) advance state-of-the-art computational methods for the long-term environmental assessment of circularity- and sustainability-transition scenarios at the scale of built districts ; (3) apply both developments to case-studies of neighborhoods in Switzerland and draw conclusions for policy-makers.

Agricultural systems are under pressure to produce increasing amounts of food for a growing human population while preserving, and ideally enhancing, the capacity of soils to sequester carbon and provide a habitat for biodiversity. Regenerative agricultural practices can help us achieve this goal. One such practice is the application of organic fertilizers. The PhD candidate will conduct a field study (in canton Vaud and Bern) to investigate the effects of organic fertilization strategies on soil greenhouse gas emissions, crop productivity, soil health, and carbon sequestration capacity. The work will be carried out in collaboration with the Nestlé institute of Agricultural Sciences.

We plan for a small team consisting of a Post-Doc and several PhD students to use and develop various novel experimental nonlinear optical scattering and imaging methods to achieve this. These methods will be applied to (1) simple 3D electrolyte in water solutions, (2) 2D electrolyte in water solutions and (3) water-ion-macromolecule systems. Linking all datasets will allow us to map both short – and long-range interactions and collective effects pertaining to all molecular actors. This allows to solve the puzzle surrounding concentrated electrolyte solutions and how they interact with macromolecules. In a well-established laboratory, the PhD student will explore next-generation bioanalytical devices operating within the infrared band of the electromagnetic spectrum for chemical specific & conformational sensitive detection of biomolecules, biological and chemical samples. PhD work will exploit novel concepts including surface enhanced infrared absorption spectroscopy (SEIRA) enabled by nanophotonic metasurfaces, hyperspectral imaging, state-of-the-art infrared equipment such as quantum cascade lasers and focal-plane array detectors as well as AI-based data science techniques. We expect that newly developed sensors will find numerous applications such as for the study and diagnosis of diseases triggered by protein misfolding as in neurodegenerative disorders, drug discovery and safety. In the Dynamic Quantum Materials Laboratory we generate materials with properties that can be manipulated on a femtosecond time scale. By driving solids with intense ultrashort laser pulses, we directly address collective modes and operate at the fundamental timescales governing the dynamics in a material. These light-matter hybrids with tunable properties opens up new ways to understand how order forms in a quantum many-body system.
Our research lies at the interface of quantum science, ultrafast laser optics and materials engineering.
