(04) Postdoctoral Fellowship Position

Postdoc Fellowship Position summary/title: Postdoctoral researcher position in Cell replacement therapy for Parkinson’s disease

We look for two postdoctoral fellows interested in developing precision cell replacement strategies for Parkinson’s disease based on cell engineering. Our laboratory has previously provided proof of concept that the conversion of somatic cells into midbrain dopamine neurons in vivo can rescue motor deficits in a mouse model of PD (Rivetti di val Cervo, 2017, Nature Biotechnology). Expanding on our previous single cell RNA-seqeuncing analysis of the developing human midbrain (La Manno et al., 2016, Cell), we have recently identified a number of factors important for the development and maintenance of adult DA neuron subtype identity (bioRxiv 2022.10.24.513487 and 2022.10.12.511898). We now aim at engineering somatic cells into subtype-specific DA neurons in order to develop more precise cell replacement strategies for Parkinson’s disease.

We are now developing this treatment for human application and are also investigating other possibilities for modifying the functions of TAMs for treatment purposes.

We aim to uncover the principles of cell fate regulation during development and repurpose them for regeneration. Using single cell multiomics, we recently discovered a latent regenerative potential in adult spinal cord stem cells and designed a targeted intervention to elicit such potential. Pneumococci are a major contributor to both morbidity and mortality and being the most common causes of milder respiratory tract infections such as otitis media and sinusitis as well as of severe diseases such as pneumonia, septicemia and meningitis. However, pneumococci are also common colonizers of the upper respiratory tract of healthy children. It is still not totally clear what determines whether the bacteria will cause a disease or just stay colonizing healthy individuals. The candidate will focus on investigation the dynamic interaction between the transcriptome and the proteome, and how the disruption of those interaction can be used to modulate gene expression and cellular phenotypes in health and disease. The candidate is expected to contribute his/her own ideas to the common goal of understanding how RNA biology and proteostasis contributes to the appearance of divergent cellular phenotypes. The candidate will perform original research in genomics, and acquire the required experimental and computational skills necessary for the completion of the project. The candidate will use varied model organism, from budding yeast to human. The candidate will be responsible for integrating different kinds of biological information to answer relevant questions (i.e. genomics, proteomics, biochemical…). The successful candidate will take a strong lead in the experimental design, and collaborate with other team members. The candidate is expected to communicate the scientific results by writing up scientific papers and attending scientific meetings. The ideal candidate is also expected to participate in the general duties of the team and to effectively communicate with scientists of very diverse backgrounds in a highly interdisciplinary and international environment.
