Phosphate and calcium are essential minerals and important constituents of the skeleton. Phosphate and calcium must be absorbed from diet, redistributed between different organs, and are either excreted into urine or reabsorbed by kidneys. These processes are controlled by transporters, hormones and metabolic factors and are important for bone health as well as for protection from uncontrolled mineralization (e.g. in the cardiovascular system or kidney stones). Aging is associated with changes in mineral metabolism, as well as with bone-remodeling, bone-loss and increased risk for renal and cardiovascular disease and death. However, the links between aging and all these processes are unclear.

The prospective PhD project aims to investigate the potential of thermal infrared remote sensing for the cryosphere. The cryosphere is changing at unprecedented rate and calls for holistic and innovative monitoring strategies to make sound prediction of future dynamics, occurrence and linked processes. This project aims to assess the spatio-temporal variability of surface temperature and linked energy fluxes over cryosphere components (snow, glaciers, permafrost) in complex, alpine topography, and prepare for the product retrieval of upcoming thermal missions (Trishna, SBG, LSTM). Specific tasks include the creation of a multi-scale dataset using intensive instrumentation, as well as across-scale post-processing and analysis of the obtained data.

Neuroscience research is among the single largest animal research fields, with around 1.3 million used each year in the EU alone. In Switzerland, more than a quarter of experimental animal used in recent years were in neuroscience, and many undergoing experimental procedures of high severity. Despite the use of these numerous experimental animals, the overall success rate in therapy development for neurological diseases such as Alzheimer’s dementia and stroke is low compared to other fields. Bridging this translational gap is critical to advancing both science and the 3Rs of replacement, reduction, and refinement. Although there are different reasons for this gap, weak or inappropriate design of preclinical studies has been flagged as key driver. For instance, the selection of animal disease models is often made based on the resources available to that researcher rather than which model provides the best representation of the specific pathophysiological process under study. Yet there is a lack of a comprehensive resource to assist preclinical neuroscientists make informed decisions on experimental parameters during study planning. This costs many experimental animals with only modest translational relevance. The advent of complex cell culture models, such as 3D organoids or microphysiological systems, has brought prospects to partly replace animal experiments for drug testing. However, no systematic curation of the application of such models has demonstrated their translational value in comparison with respective animal models.

In this project, a systematic computation of long-term series of rock glacier kinematics (horizontal velocities and vertical changes) will be compiled for a large number of sites located in various topo-climatic settings. We are interested in Hemophagocytic Lymphohistiocytosis (HLH), a group of genetic disorders of lymphocyte cytotoxicity predisposing children and infants to severe multi-system inflammatory syndrome. Immune stimulation, e.g. following infections, is poorly controlled, leading to excessive activation of T cells and macrophages and a life threatening cytokine storm. Current treatment, which aims at eliminating or suppressing activated immune cells, followed by hematopoietic stem cell transplantation, is still associated with a mortality of around 30-40%. Recently, more targeted therapies directed at neutralisation of cytokines, for which we have performed decisive preclinical studies, have been evaluated, but it remains unclear whether they will be sufficient to timely and reliably induce complete remission. Our aim is to better characterise the molecular pathogenesis of the disease and find novel targetable factors that can tip the balance away from excessive immune stimulation. We aim to do so in an interdisciplinary manner by combining genomics, high-content imaging, use of repurposed drug libraries, and disease modelling in iPSCs or mouse models.

PhD position summary/title: PhD Student Position in Time-Resolved Studies of Protein Dynamics

Project description: The research is centered around the exciting new possibilities in time-resolved structural biology offered by the Swiss Light Source and the Swiss X-ray free-electron laser, investigating the allosteric signaling between domains in various photoactive protein complexes. These facilities allow the determination of 3D structures of transient protein intermediates with atomistic spatial and ultrafast (up to 100 fs) temporal resolution. Time-resolved IR spectroscopy will be used in the project to specify selected time points, at which the population of intermediates is maximized and their 3D structures can be determined. Quantum chemical and molecular dynamics simulations based on these 3D structures should cross-validate the experimental IR spectra. Final cell biology experiments will close the loop from the initial stimulation to signaling output.
