Postdoc Fellowship Position summary/title: Computational Genomics Research Associate

A new position is available for an experienced post-doctoral Research Associate in Computational Genomics in the UK Dementia Research Institute (UK DRI) research group led by Dr Sarah Marzi.

You will lead one or more projects characterising the epigenome and transcriptome of brain cell types in the context of neurodegenerative diseases, including Alzheimer’s disease (AD), Parkinson’s disease and ALS.  You will be based at King’s College London under the supervision of Dr Sarah Marzi (https://www.kcl.ac.uk/people/sarah-marzi). You will contribute to the design and analysis of large-scale ‘omics datasets with a primary focus on analysis of the epigenome and chromatin architecture to address major research questions relevant to AD.

This opportunity would suit a bright, talented, highly motivated, and ambitious individual to lead one or more computational projects exploring the transcriptome and epigenome of brain cell types relevant to disease. The post holder will use next-generation sequencing data from patient-derived brain tissue and stem cell model systems to identify signalling pathways and transcription factors dysregulated in brain diseases with a focus on Alzheimer’s disease. You will use genomic datasets that may include (long reads) RNA-seq, ChIP-seq, CUT&Tag, ATAC-seq and single cell/nuclei datasets. These studies will help to guide the functional interpretation of disease risk variants and environmental risk factors for aging-related brain disorders such as Alzheimer’s disease and Parkinson’s disease.

Postdoc Fellowship Position summary/title: Research Associate in Human Autonomic Cardiorespiratory Control

This department has a focus on fundamental questions regarding human physiological function and adaptation in health and disease and occupies an important nexus between basic biomedical sciences, clinical and applied research. It comprises four interlinking research groupings operating under an overarching strategy focused on furthering understanding of integrative human physiology. Our research encompasses a unique whole-body systems through to stem cell approaches which is aimed at gaining insight into factors affecting human health, disease, biological ageing and exercise, as well as performance in unique and extreme environments. We are committed to integrative and translational research which is facilitated by collaborations with other research groupings within King’s College London, King’s Health Partners, industry and the military.

Postdoc Fellowship Position summary/title: Research Associate – Immunology

Unlike in hematologic malignancies, the efficacy of chimeric antigen receptor (CAR) modified T cells in solid tumors such as breast cancer has been very limited.  Oncolytic viruses (OVs) encoding a variety of transgenes have been evaluated as therapeutic tools to increase the efficacy of CAR-modified T cells within the typically immunosuppressive solid tumour microenvironment (TME). Using fully systemic delivery of OVs and CAR T in fully immunocompetent mouse models, we have defined a new mechanism by which OVs can potentiate CAR T efficacy against solid tumours.  Our novel approach shows that stimulation of the native T cell receptor (TCR) in the CAR T cells against either OV-derived viral antigens, or against OV-encoded tumour antigens, gives rise to enhanced proliferative and functional properties and distinct memory phenotypes of the CAR T. In vivo expansion of dual specific (DS) CAR T (specific for both CAR target antigen through the CAR and viral/tumour antigen through the TCR) was leveraged by in vitro pre-loading with oncolytic Vesicular Stomatitis virus (VSV) or reovirus.  This allowed for a further in vivo expansion/re-activation by homologous boosting, which led to high cure rates in murine models of subcutaneous melanoma and intra-cranial High-Grade Glioma (HGG) (Evgin et al., 2022 Science Translational Medicine).  We now plan to test this strategy pre-clinically in the treatment of breast cancer leading directly to the generation of IND-enabling data to translate DS CAR T cell therapy into a fully systemic protocol for CAR T/OV combination therapy against breast tumours which, critically, does not require direct access to the tumour for administration.

Postdoc Fellowship Position summary/title: Post Doctoral Research Associate in sub-cellular mass spectrometry

The purpose of this role is to develop new mass spectrometry techniques that can be used to enable users of the facility to make new measurements of material sampled using the SEISMIC facility.  Analysis of single-cells using mass spectrometry is challenging, and yet analysis of sub-cellular compartments is the next frontier, possible at SEISMIC and in high demand by biologists.  Kings have recently made a significant investment in mass spectrometry instrumentation, to enable the development of this approach by capillary sampling and mass spectrometry. 

The post holder will work with the SEISMIC team and KCL core facilities team to develop new mass spectrometry techniques that can be used to analyse capillary sampled cells and their sub-cellular compartments.  They will be encouraged to present their work at national and international conferences, and work with users of the facility to demonstrate proof of concept applications.  They will be encouraged to submit applications for funding, for example for fellowships to support their future career.

We are seeking a Postdoctoral Research Associate to join the Faculty of Cardiovascular Medicine & Sciences, the core constituent of the BHF Centre.

The applicant will work on a Diabetes UK-funded project aiming to uncover novel events in diabetic microangiopathy that can be targeted pharmacologically to restore the function of the microvasculature. Microangiopathy is a major complication in diabetes mellitus with a pronounced impact on microvascular perfusion, vessel rarefaction and tissue ischemia. Using human blood vessel organoids (BVOs) we recently demonstrated that they capture key aspects of the microvasculature and respond rapidly to metabolic rewiring (Romeo et al Nat Commun 2023  10.1038/s41467-023-41326-2). In this project, the successful applicant will utilize diabetic patient-derived BVOs to establish a functional screening platform that will enable personalized therapeutic approaches. This pipeline will serve as a personalized system to test existing and new medications to identify effective drugs for each diabetic patient.

The McTernan Group aims to apply recent breakthroughs in artificial molecular machines and metal-organic capsules in biologically relevant settings. We work with rotaxanes, catenanes and capsules to synthesise functional architectures, creating de novo catalytic enzyme analogues, artificial cellular receptors, and generating targeted drug delivery vehicles. We work both in basic science, and translational applications.

This the successful candidate for this 24-month role will lead our chemical biology research programme; designing and conducting experiments, liaising with world-leading STPs at the Francis Crick Institute, developing and deploying assays, training junior researchers, and liaising with CROs. They will take the lead on assaying the biological activity of the numerous therapeutic molecules developed in our laboratory, and help guide their successful optimisation, working closely with chemists across our research portfolio.
