The post holder will work within a programme focused on mitochondrial transport proteins, which are crucial for the functioning of mammalian cells. This project is funded by Apollo Therapeutics and focusses on the development of specific inhibitors of the mitochondrial pyruvate carrier (MPC). MPC has emerged as a promising drug target for metabolic disorders, including non-alcoholic steatohepatitis and diabetes, metabolically dependent cancers and neurodegenerative diseases. The human MPC is a heterodimer of two homologous small membrane proteins, forming two distinct complexes either MPC1/MPC2 or MPC1L/MPC2 [1]. The goal is to develop novel inhibitors of human MPC to treat the aforementioned diseases. Thermostability shift and transport assays have already been developed to screen for better and more specific binders [1]. The project involves a collaboration with Apollo and associated Contract Research Organizations as well as with different experts on mitochondrial biology within the MRC-MBU. The post-holder will participate in every stage of the process, including protein purification, screening of novel binders by thermostability shift and transport assays, interacting with other team members and collaborators, and preparing of manuscripts/IP.

We seek a statistician interested in human genetics who wants to apply machine learning to estimate cell population frequencies from bulk gene expression data. A common approach to identify dysregulated pathways in diseases is to compare gene expression profiles from healthy vs disease samples. However, the cellular composition of many biological samples is heterogeneous, and, given that many disease processes involve cell infiltration and expansion, such comparisons are confounded by changes in cellular composition. In addition, it is generally recognised that rare populations play a critical role in the pathogenesis of complex diseases, therefore, the ability to measure and interpret phenotypic changes between specific conditions at the cell subset level is critical to obtain a detailed understanding of the role of each cell subset in disease. However, in practice, measuring the frequency of such populations in large cohorts of human samples can be very challenging. Thus, methodologies to infer population frequencies from bulk transcriptome data opens up many possibilities towards understanding disease mechanisms.

The beneficial interaction of plants with endomycorrhizal fungi commences via mutual recognition in the rhizosphere. We identified a receptor complex that is required for perception of beneficial fungi by plants [Gutjahr, Gobbato et al., Science (2015), Choi et al., Nat Commun 2020], and also for plant development. Preliminary data suggest that the symbiosis signalling ability is a derived and not an ancestral trait. The post holder will work within a programme focused on mitochondrial transport proteins, which are crucial for the functioning of mammalian cells. This project is funded by Apollo Therapeutics and focusses on the development of specific inhibitors of the mitochondrial pyruvate carrier (MPC). MPC has emerged as a promising drug target for metabolic disorders, including non-alcoholic steatohepatitis and diabetes, metabolically dependent cancers and neurodegenerative diseases. The human MPC is a heterodimer of two homologous small membrane proteins, forming two distinct complexes either MPC1/MPC2 or MPC1L/MPC2 [1]. The goal is to develop novel inhibitors of human MPC to treat the aforementioned diseases. Thermostability shift and transport assays have already been developed to screen for better and more specific binders [1]. The project involves a collaboration with Apollo and associated Contract Research Organizations as well as with different experts on mitochondrial biology within the MRC-MBU. The post-holder will participate in every stage of the process, including yeast culturing, protein purification, drug screening procedures, and they will be interacting with other Unit members and collaborators.
